Immunocytochemistry/Immunofluorescence: Tenascin C Antibody (4C8MS) [NB110-68136] - SK-MEL-28 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-Tenascin C at 5 ug/ml overnight at 4C and detected with an anti-Mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry-Paraffin: Tenascin C Antibody (4C8MS) [NB110-68136] - IHC analysis of a formalin fixed paraffin embedded tissue section of mouse bone-tendon using Tenascin C antibody (clone 4C8MS) at 1:25 dilution. The signal was detected using HRP-DAB detection method which followed counterstaining using hematoxylin. The antibody generated a very specific cytoplasmic, membrane and extra-cellular signal in tendon fibroblasts, osteoblasts, osteoclasts, and some bone marrow cells. The mineralized areas were largely negative for the staining.

Flow Cytometry: Tenascin C Antibody (4C8MS) - BSA Free [NB110-68136] - An intracellular stain was performed on MCF7 cells with Tenascin C Antibody (4C8MS) NB110-68136 (blue) and a matched isotype control MAB002 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (84540, Thermo Fisher).

• Background: Tenascin C is an extracellular matrix protein that is expressed in various tissues during development, disease or injury. Tenascin C inhibits HIV infection in immune cells by binding to a chemokine coreceptor site on the HIV-1 envelope protein, thereby blocking the virus' entry into the host cells. Background

Tenascin, also known as hexabrachion and cytotactin, is an extracellular matrix protein with a spatially and temporally restricted tissue distribution. It is a hexameric, multidomain protein with disulfide linked subunits of 190 to 240 kD, originally characterized as 'myotendinous antigen.' In the embryo it is present in dense mesenchyme surrounding developing epithelia and in developing cartilage and bone. In the adult, tenascin remains present in tendons and myotendinous junctions in the perichondrium and periosteum, as well as in smooth muscle. Tenascin (TN)(Erickson and Bourdon 1989, Erickson 1993) is a high molecular weight, multifunctional, extracellular matrix glycoprotein, expressed in association with mesenchymal-epithelial interactions during development and in the neovasculature and stroma of undifferentiated tumors. It has been described under a variety of names: cytotactin, hexabrachion protein, J1-200/220, myotendinous antigen (MI), neuronectin (NEC1) and glioma mesenchymal extracellular matrix (GMEM). The TN molecule is a disulfide-linked hexamer. Human TN has 3 subunits of 190, 200 and 220 kDa. It is primarily made up of 14.5 epidermal growth-factor-like repeats, 15 units similar to the fibronectin type-III-homology repeat and, at the C-terminus, has a sequence with homology to the globular domain of the beta and gamma chain of fibrinogen. Monoclonal antibody reacting specifically with tenascin is an essential tool for the localization, identification and studies on the role of the molecule in epithelial-mesenchymal and neuronal-glial interactions (Castellucci et al. 1991, Balza et al. 1993).
