Immunocytochemistry/Immunofluorescence: TIPIN Antibody [NBP2-98947] - Immunofluorescence staining of TIPIN in HeLa cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with NBP2-98947 antibody (dilution ratio 1:200) at 4C overnight. Then cells were stained with the Alexa Fluor(R)488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus and cytoplasm.

Background

Timeless-interacting protein (TIPIN) was originally isolated as a protein that interacts with timeless, a factor that plays a role in circadian rhythm and cell cycle checkpoint regulation. The TIM-TIPIN complex has been shown to coordinate the intra-S-phase checkpoint in response to UV-induced DNA damage. TIPIN is required for cell cycle arrest in response to DNA damage. It also interacts with RPA (replication protein A) on DNA and is able to slow down DNA chain elongation in response to UV-damage.
