Background
Spectrin beta 3 (SPTBN2) belongs to spectrins family of proteins which are essential to mechanical support/structural membrane integrity maintenance in erythrocytes, stabilizing cell-cell contacts, and localizing ion channels as well as cell adhesion molecules within specific subdomains of plasma membranes. Vertebrate spectrins have two alpha-subunits (alpha-I/alpha-II), four beta-subunits (beta-I-beta-IV) and a beta-H subunit creating diversity and specialization of function. Spectrin beta 3 is primarily expressed in nervous tissues with highest expression levels in the cerebellum, where it is found in Purkinje cell soma and dendrites. Spectrin beta 3 interacts with EAAT4, the glutamate transporter predominately expressed in Purkinje cells, and stabilizes it at the plasma membrane leading to glutamate clearance from the synaptic cleft, and resulting in both modulation of glutamatergic neurotransmission as well as prevention of glutamate-mediated neurotoxicity. Defective spectrin beta 3 has been established as causing factor in spinocerebellar ataxia type 5 (SCA5).

Do you have stability tests done on NB110-58346 if left at RT for several hours ? Customer is doing automatic staining protocol where the antibody needs to stay several hours at RT and they do not get the nice results as with a manual staining, so we are wondering if the antibody would inactivate or get aggregated?
Unfortunately, we do not have any stability data on Spectrin beta 3 antibody with catalogue number NB110-58346, however, typical staining protocols involve a couple of hours incubation at room temperature or are performed overnight at 4C. Therefore, I am assuming that the customer should be OK by incubating tissues sections with NB110-58346 for little over a couple of hours without any problems. It may be noted however that the extended incubation with primary antibodies may lead to non-specific binding or overstaining.
