Background
ROR gamma/RORC/NR1F3, aka RAR Orphan Receptor gamma, is a Nuclear Receptor 1 Thyroid Hormone-Like receptor containing a short A-T rich sequence follow by a single core motif half-site 5'-AGGTCA-3'. RORC is a key regulator of thymopoiesis, bone metabolism, T-cell apoptosis, and lymphoid organogenesis. NR1F3 regulates intrinsic transcriptional functions. Unlike other major nuclear receptors, RORC binds as a monomer to hormone response elements and is documented in mouse thymus, adipose, bone, skeletal muscle, liver, kidney, and human skeletal muscle. An N-terminal isoform of ROR gamma, ROR gamma T, inhibits Fas ligand expression and cytokine secretion in immature thymocytes.

Since the protocol to isolate cells from mice seems to be quite long I would like to know if you have experience in staining with Foxp3 and Ror gamma and leaving cells overnight? Would you suggest leaving cells overnight in Fix/Perm or better after having completed the staining? And in which buffer would you leave them then?
Where as FOXP3 and ROR gamma staining can be stopped after the fixation step, we suggest that researchers strictly follow the protocol without variation (even if it takes a long time), when trying for the 1st time. After you establish this for your system, then you can work on optimizing a varied protocol according to your needs and convenience. In this case, after the fix/perm step (Step 13 of NBP2-266911) wash the cells in 1X Staining buffer (2 times) and suspend in Staining buffer (500 ul) overnight. Then, in the morning proceed to Step 14 and continue the protocol.
