Background
RNA polymerase II catalyzes DNA transcription into RNA, and is composed of 12 subunits. The largest of these subunits is B1 (RPB1, POLR2A), which contains the DNA binding domain and the enzymatic subunit. RPB1 contains multiple heptapeptide repeats in the CTD arranged in the sequence Tyr1-Ser2-Pro3-Thr4-Ser5-Pro6-Ser7 (YSPTSPS). Modifications of this sequence, such as phosphorylation, alter polymerase activity. Serines 2, 5, and 7 are known to become phosphorylated to lead the complex through the transcription cycle, as well as functions in polyadenylation, mRNA capping, and snRNA-processing.
