Background
The structure and integrity of DNA molecules can be spontaneously affected through intrinsic instability of chemical bonds in DNA or can be induced by chemical compounds and irradiation. DNA lesions are of many different types including single and double-strand breaks, inter and intra-strand cross-links and different kinds of base modification. DNA lesions hamper processes like transcription and replication resulting in cell cycle arrest, apoptosis and genomic instability (mutagenesis). DNA lesions have been implicated in many distinct genetically inherited diseases, in carcinogenesis, origin of genetic disorders and in aging. Rad1 is an essential component of cell cycle checkpoints activated by DNA damage and incomplete DNA replication. Six Rad proteins (Rad1, Rad3, Rad9, Rad17, Rad26, and Hus1) are involved in the early steps of DNA damage checkpoint response in S. pombe. Human HRAD1 gene is located on chromosome 5p13.
