PMP22 Antibody (JM52-30)

Western Blot: PMP22 Antibody (JM52-30) [NBP2-67068] - Western blot analysis of PMP22 on PC-12 cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1:5,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: PMP22 Antibody (JM52-30) [NBP2-67068] - Staining PMP22 in N2A cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: PMP22 Antibody (JM52-30) [NBP2-67068] - Immunohistochemical analysis of paraffin-embedded human pancreas tissue with Rabbit anti-PMP22 antibody at 1/500 dilution.The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/500 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: PMP22 Antibody (JM52-30) [NBP2-67068] - Analysis of SH-SY5Y cells with PMP22 antibody at 1/100 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black).

Immunohistochemistry-Paraffin: PMP22 Antibody (JM52-30) [NBP2-67068] - Analysis of paraffin-embedded rat brain tissue using anti-PMP22 antibody. Counter stained with hematoxylin.

Background

Peripheral Myelin Protein 22 (PMP22) is a 160 amino acid glycoprotein which is a member of the claudin family and is thought to play an important role in the structure of peripheral nerve myelin. Irregularities in its gene results in hereditary demyelinating neuropathies. PMP22 protein is present in the compact myelin of adult human peripheral nerve and its distribution mimics that of Myelin Protein zero (P0).
