Notes

Purification Notes After repeated injections, immune eggs were collected, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using a peptide column, and the concentrations of the eluates adjusted to 200 ug/ml. Finally, equal volumes of both of these affinity purified anti-peptide antibodies were mixed, and the preparation was filter-sterilized. Storage Notes Store at 4C in the dark. The antibody should have a shelf life of at least 12 months (provided it is kept under sterile conditions). Do not freeze this antibody unless you want to store it for longer periods of time. Note, however, that each time an antibody preparation is frozen, about half its binding activity is lost.

Background

Myelin Protein Zero encodes a major structural protein of peripheral myelin. Mutations in this gene result in the autosomal dominant form of Charcot-Marie-Tooth disease type 1 as other polyneuropathies. [provided by RefSeq]

  1. Does this antibody target the extracellular domain of the protein?
    • Two different peptides were used to raise this antibody: 1) the first peptide sequence is located in the amino-terminal half of the protein and is located within an "Immunoglobulin V-type" domain, which is extracellular. 2) The second peptide sequence is located near the C-terminus of the protein (cytoplasmic). The antibody recognizes MPZ protein, when both antigens are available but we do not know how strong would be the signal if only extracellular antigen is visible.
