Background

Human Myelin Protein Zero (Po) is a 28 kDa protein (258 amino acids) found in myelin of the PNS. Expressed in myelinating Schwann cells, Po serves as a homophilic adhesion molecule, allowing adjacent laminae of the myelin sheath to undergo compaction. Po is also a marker of a subpopulation of neural crest cells. Mutations of the MPZ gene product in humans is responsible for Charcot-Marie-Tooth type 1B neuropathies (HMSN1B).

  1. Does this antibody target the extracellular domain of the protein?
    • Two different peptides were used to raise this antibody: 1) the first peptide sequence is located in the amino-terminal half of the protein and is located within an "Immunoglobulin V-type" domain, which is extracellular. 2) The second peptide sequence is located near the C-terminus of the protein (cytoplasmic). The antibody recognizes MPZ protein, when both antigens are available but we do not know how strong would be the signal if only extracellular antigen is visible.
