Western Blot: PDIA2 Antibody [NB100-1921] - Normal human brain.

Immunocytochemistry/Immunofluorescence: PDIA2 Antibody [NB100-1921] - IF Confocal analysis of 3T3 cells using PDIA2 antibody (NB100-1921, 1:5). An Alexa Fluor 488-conjugated Goat to rabbit IgG was used as secondary antibody (green). Actin filaments were labeled with Alexa Fluor 568 phalloidin (red). DAPI was used to stain the cell nuclei (blue).

Immunohistochemistry-Paraffin: PDIA2 Antibody [NB100-1921] - IHC analysis of a formalin fixed paraffin-embedded (FFPE) human brain using 1:250 conc. of PDIA2 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Cytoplasmic staining of PDIA2 in neurons was observed. Staining was performed by Histowiz.

Immunocytochemistry/Immunofluorescence: PDIA2 Antibody [NB100-1921] - PDIA2 staining in Hela cells detected with a Dylight 488 labeled secondary antibody (Green) with Hoechst 33258 nuclear counterstain (Blue).

Background

This gene encodes a member of the disulfide isomerase (PDI) family of endoplasmic reticulum (ER) proteins that catalyze protein folding and thiol-disulfide interchange reactions. The encoded protein has an N-terminal ER-signal sequence, two catalytically active thioredoxin (TRX) domains, two TRX-like domains and a C-terminal ER-retention sequence. The protein plays a role in the folding of nascent proteins in the endoplasmic reticulum by forming disulfide bonds through its thiol isomerase, oxidase, and reductase activity. The encoded protein also possesses estradiol-binding activity and can modulate intracellular estradiol levels.
