Application Notes
A protein band of ~36 kDa is detected in mouse lung tissue lysate. Pre-incubation with the immunizing peptide completely removes langerin reactivity from NB100-56733. Use in Flow-intracellular reported in scientific literature (PMID: 18369922).

Background

Langerin (CD207) is a type II membrane-associated C-type lectin known to be expressed exclusively by Langerhans cells. Langerin recognizes mannose residues via its single carbohydrate recognition domain (CRD). Langerin is localized not only on the cell surface, but also intracellularly in close association with Birbeck granules. Transfection of Langerin cDNA into fibroblasts creates a compact network of membrane structures with typical features of Birbeck granules (BG). Langerin is thus a potent inducer of membrane superimposition and zippering leading to BG formation.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: NOD2 Antibody (2D9) [NB100-524] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-NOD2 (2D9) NB100-524 at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Rat brain neural cultures stained with NBP1-92684 (green), a chicken polyclonal antibody to GFAP (NB300-213, red) and DNA (blue). The NBP1-92684 antibody reveals strong cytoplasmic staining in a population of small developing neurons. These cells are often found in small clumps as in these cultures, as in this example. Note that they are not positive for MAP2, which is characteristic of more mature neurons, and that the mature neuron shown is negative for NBP1-92684. This Doublecortin antibody is therefore an excellent marker of developing neuronal cells.

  1. I’m contacting you because I’m using often your Antibodies and I would like to conjugate some of them by myself. For that purpose I would need to have Antibodies in a buffer solution that do not contains any additive like NaAz or BSA. Could you please tell me if it is possible to order such antibodies, and if there is a minimum volume quantity to order?
    • Typically there is a minimum order quantity for custom formats of our products (1mg on average). However we are transitioning to offering more of our products in an azide/BSA free format. Please let us know which products are needed, and we can check the feasibility of offering them in the desired format.
  2. Would you please recommend a dilution for this application? Do you have any suggested protocol for IHC?-EndFragment-->
    • For IHC-P we recommend a dilution range of 1:100-1:500. The lab used the following protocol: http://www.novusbio.com/support/protocols/immunohistochemistry-paraffin-protocol.html.
