- Assay Diluent A (Item D): 30 ml of animal serum with 0.09% sodium azide as preservative. ForStandard/Sample (serum/plasma) diluent.
- Assay Diluent B (Item E): 15 ml of 5x concentrated buffer. For Standard/Sample (cell culturemedium/urine) diluent.
- Detection Antibody IL-9 (Item F): 2 vial of biotinylated anti- mouse IL-9 (each vial is enough to assay half microplate).
- HRP-Streptavidin Concentrate (Item G): 8 µl 70,000x concentrated HRP-conjugated streptavidin.
- IL-9 Microplate (Item A): 96 wells (12 strips x 8 wells) coated with anti-mouse IL-9.
- Standards (Item C): 2 vials, recombinant mouse IL-9.
- Stop Solution (Item I): 8 ml of 2 M sulfuric acid.
- TMB One-Step Substrate Reagent (Item H): 12 ml of 3,3’,5,5’-tetramethylbenzidine (TMB) in buffered solution.
- Wash Buffer Concentrate (20x) (Item B): 25 ml of 20x concentrated solution.

Background
IL-9 is a cytokine with immunoregulatory function, produced by activated T-helper type 2 lymphocytes. It acts on various cell types such as T- and B-lymphocytes, mast cells, haemopoietic progenitors and megakaryoblastic leukemia cell lines. IL-9 stimulates cell proliferation and prevents apoptosis. IL-9 may play a role in the pathogenesis of diseases such as certain lymphomas (Hodgin's disease, anaplastic lymphoma) and asthma. Although murine IL-9 can act on human cells, human IL-9 is inactive on mouse cells.
