Western Blot: CD48/SLAMF2 Antibody (JE37-98) [NBP3-32157] - Western blot analysis of CD48/SLAMF2 on different lysates with Rabbit anti-CD48/SLAMF2 antibody (NBP3-32157) at 1/1,000 dilution. Lane 1: Raji cell lysate Lane 2: Jurkat cell lysate Lysates/proteins at 15 ug/Lane. Predicted band size: 27.7 kDa Observed band size: 40 kDa Exposure time: 80 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32157) at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:100,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: CD48/SLAMF2 Antibody (JE37-98) [NBP3-32157] - Immunohistochemical analysis of paraffin-embedded human colon tissue with Rabbit anti-CD48/SLAMF2 antibody (NBP3-32157) at 1/1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32157) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

CD48 is a glycosyl phophatidyl inositol (GPI) anchored cell surface protein also known as Blast1, HuLy m3, BCM1 (in mouse), and OX45 (in rat). CD48 is strongly expressed on lymphocytes and monocytes and weakly on granulocytes but is absent on platelets, fibrolasts, epithelium and endothelium. CD48 is one of the markers for detecting the defects of GPI anchoring structure on patients with paroxysmal nocturnal hemoglobulinuria (PNH) and serves as a low affinity ligand for CD2. The CD48 antigen is closely related to the activation antigen, Blast 1.
