Background

The CD19 antigen (95kDa) is expressed from the earliest stage of B progenitor development, on all peripheral B cells including germinal centre B cells, and all B cell lines and B cell leukaemia tested. T cell and monocytic cell lines are negative and the antigen is lost on B cell maturation to plasma cells. The antigen is a type I integral membrane glycoprotein whose in vitro inhibition will influence B cell activation and proliferation.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: NOD2 Antibody (2D9) [NB100-524] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-NOD2 (2D9) NB100-524 at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western blot shows lysates of LNCaP human prostate cancer cell line and PC‑3 human prostate cancer cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human Neprilysin/CD10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1182) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). For additional reference, Recombinant Mouse Neprilysin (Catalog # 1126-ZN) (5 and 50 ng/lane) and Recombinant Human Neprilysin (CHO-expressed) (Catalog # 1182-ZNC) (5 and 50 ng/lane) were included. Specific bands were detected for Neprilysin/CD10 at approximately 100 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Neprilysin/CD10 was detected in immersion fixed paraffin-embedded sections of human kidney using Goat Anti-Human Neprilysin/CD10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1182) at 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue).
