Human Caspase 9 recombinant protein catalytic subunits (NP_001220).
Specificity
This antibody detects both the pro- and active-Caspase 9 forms. In western blots, the proform of caspase-9 is detected at ~50 kDa, the large subunit at ~35 kDa, and the small subunit at ~15 kDa. Intermediate caspase-9 cleavage forms may also be seen at ~21 kDa.

Application Notes
Caspase-9 immunostaining may appear diffuse or punctate. It may be localized in the cytosol, mitochondria, and nucleus. Caspase-9 staining in the nucleus is considered to be an indication of active Caspase-9. Both pro and active/cleaved Caspase-9 staining may also be seen in the mitochondria. 2. Caspase antibodies are classical tools for detecting inactive (pro) and active (cleaved) forms of the enzymes. The presence of the large or small subunits in western blots is considered to be a marker of caspase activation. Use of this antibody in Flow-Intracellular reported in scientific literature (PMID 24804954)

Background

Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. A novel member in the caspase family was recently identified and designated ICE-LAP6, Mch6, and Apaf-3. Caspase-9 and Apaf-1 bind to each other, which leads to caspase-9 activation. Caspase-9 is also activated by granzyme B and CPP32. Activated caspase-9 cleaves and activates caspase-3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis. Caspase-9 play a central role in cell death induced by a wide variety of apoptosis activators including TNFalpha, TRAIL, anti-CD-95, FADD, and TRADD. Caspase-9 is expressed in a variety of human tissues.

  1. Could I use HeLa cell lysate as a positive control for Caspase 9 primary antibodies in WB?
    • After looking through our testing it does appear you should be fine using HeLa as a positive control cell lysate for Caspase 9 in Western Blot, as we have used this sample successfully in our QC tests and shown expression of Caspase 9.
