Immunogen
Phospho-AIF (Ser-116) synthetic peptide (coupled to carrier) corresponding to amino acids surrounding Ser-116 in human AIF. This site is conserved in rat and mouse AIF, but the amino acids surrounding the site are not well conserved. The site is not found in AIFM2 and AIFM3. [UniProt# O95831]

Specificity
The antibody was cross adsorbed to unphosphorylated AIF (Ser-116) peptide before affinity purification using phospho-AIF (Ser-116) peptide. This antibody antibody detects a triplet at 66, 62, and 57 kDa protein on SDS-PAGE immunoblots of human Jurkat cells treated with calyculin A. This reactivity is not observed after lambda phosphatase treatment to dephosphorylated AIF.

Background

AIF, or apoptosis-inducing factor, is a flavoprotein localized to the mitochondrial membrane that plays a primary role in caspase-independent cell death, or apoptosis (1,2). AIF translocates to the nucleus when apoptosis is induced and causes mitochondria to release the apoptogenic proteins, cytochrome c and caspase-9. Apoptotic activity of AIF leads to chromatin condensation and DNA fragmentation (1). Similar to cytochrome c, AIF is a bifunctional protein with an oxidoreductase function and a second apoptogenic function. In contrast to cytochrome c, however, AIF acts in a caspase-independent fashion (2,3,4).
