![Western Blot: Collagen I Antibody [NB600-408] - The effects of PA on TGF-beta 1-induced alpha - SMA & collagen I alpha 1 expression in HSC-T6 cells. (A,B) The inhibitory effect of PA on TGF-beta 1-induced alpha -SMA & collagen type I mRNA & protein expression in rat hepatic stellate cells. Cells were pretreated with 0.5, 1, & 2 μM PA for 1 h, & then stimulated with TGF-beta 1 (5 ng/mL) for 24 h. Total RNA extracted from cells was analyzed by the real-time polymerase chain reaction to determine alpha -SMA & ColIa1 mRNA expression; (C) the total protein extracted from cells was subjected to Western blotting to determine alpha -SMA & collagen I alpha 1 expression. Protein bands were imaged using densitometry & analyzed using ImageJ software. The relative expression levels of target proteins were normalized using beta -actin as an internal control. ![Western Blot: Collagen I alpha 1 Antibody - Azide and BSA Free [NBP1-30054] - TNC deficiency reduced kidney fibrosis in animal models.Deletion of TNC (TNC−/−) significantly attenuated the induction of collagen I assessed by IF following UUO by approximate 30% at day 7 & 10 compared with their wild-type littermates (A, n = 4 for each time point, two-way ANOVA p < 0.05). Consistently, the expression of fibrosis markers, such as collagen I alpha , fibronectin & plasminogen activator inhibitor-1 (PAI-1), were significantly lower in TNC−/− mice at UUO day 7 & 10 (B, n = 4 for each time point, two-way ANOVA, p < 0.05). Western blot showed that the proteins of collagen I alpha & alpha -SMA were also significantly reduced in TNC−/− mice at UUO day 7 (C, n = 7, p < 0.05). STA-ET-8.2 cells were FACS-sorted & CD133+ & CD133- fractions treated with increasing concentrations of (A) Doxorubicin (Doxo), (B) Etoposide (Etop), (C) Vincristine (Vinc) or (D) a combination of all three drugs at high (Doxo 10 μM; Etop 10 ug/ml; Vinc 100 ng/ml), medium (Doxo 2.5 μM; 20 Etop 2.5 ug/ml; Vinc 25 ng/ml) & low (Doxo 0.5 μM; Etop 0.5 ug/ml; Vinc5 ng/ml) concentrations. ![Immunocytochemistry: N-Cadherin Antibody (13A9) [NBP1-48309] - Human meningioma mouse xenograft model KCI-MENG1-LPSX generated with the spontaneously immortal cell line KCI-MENG1-LP. Tumors from immunocompromised SCID mice were dissected and the derivative cell line KCI-MENG1-LPSX CL was generated. The EMA, PR, and N-cadherin IHC of the mouse tumor highly resembled the original patient-derived tumor. The vimentin- and Ki-67-stained cells in the mouse tumor tissue were markedly more abundant and more intensely stained than in the original tumor. KCI-MENG1-LPSX CL cells displayed the same patterns of immunostaining as the high passage parent cell line KCI-MENG1-HP, including the loss of PR staining. Scale bar 50 um. ![Immunocytochemistry/Immunofluorescence: CXCR4 Antibody [NB100-74396] - Optimization of immunofluorescence staining of CXCR4 protein. MDA, PC3, KG1 cells were used for the optimization experiments. Cells were seeded onto cell-tak coated 48 mm coverslips in a 48-well plate. Cells were fixed with 2% formaldehyde for 20 min, washed with PBS. After fixation, cells were blocked with 2.0% BSA in PBS for 1 h at room temperature. All the cells were incubated with a primary antibody for anti-rabbit CXCR4 for 1 h. After washing with PBS, cells were put in respective secondary antibodies-anti-rabbit dylight 405 for 1 h. PSMA= Magenta, EpCAM= Yellow, sLex= Green, CXCR4= Red, and Merge shows all the colors. MDA=PSMA+, EpCAM+, sLex+, CXCR4+. PC3 = PSMA-, EpCAM+, sLex-, CXCR4+. KG1 = PSMA-, EpCAM-, sLex+, CXCR4-. CXCR4 & MYH11 expression levels in mouse PASMCs were analyzed by immunofluorescence staining (a; n = 3). CXCR4 protein is stained red, & MYH11 is stained green to indicate the PASMCs (×400; scale bars indicate 50 μm). CXCR4 fluorescence intensity was calculated (b; n = 3). CXCR4 protein expression levels in lung tissue were examined by western blot. GAPDH served as an internal control (c, d; n = 3). Data are presented as the mean ± SD. #Value significantly greater than the corresponding value in saline-treated normoxic mice (##P 50 CTCs/ 2.5 ml blood) was processed via ficoll density centrifugation & the PBMC fraction was collected. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. All prostate CTCs expressed CXCR4, while, sLex expression was variable. The analysis of sLex intensity is shown in Figure 4. ![Immunocytochemistry/Immunofluorescence: RUNX2/CBFA1 Antibody [NBP1-77461] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with RUNX2/CBFA1 Antibody (NBP1-77461) at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes.
