Western Blot: Survivin Antibody [NB500-201] - Analysis of 30ug of HeLa whole cell lysate [NB800-PC1] using rabbit polyclonal [NB500-201] at 1ug/ml. Detection was performed using ECL method with 1 minute exposure. Band detected at higher molecular weight than the predicted MW (16 kDa).

Flow Cytometry: Survivin Antibody [NB500-201] - An intracellular stain was performed on HeLa cells with [NB500-201] and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550.

Detection of collagen I in Wistar rat hepatic stellate cells (HSC) in control (GFP-transduced) (left lane) and PPARg-transduced cell lysates (right lane). Protein staining shown below each blot depicts equal protein loading. An equal amount of the whole cell protein (100 ug) was separated by SDS-PAGE and electroblotted to nitro-cellulose membranes. Proteins were detected by incubating the membrane with anti-Collagen I antibody at a concentration of 0.2-2 ug/10 mL in TBS (100 mM Tris-HCl, 0.15 M NaCl, pH 7.4) with 5% non-fat milk. Detection occurred by incubation with a HRP conjugated secondary antibody at 1 ug/10 mL. Proteins were detected by a chemiluminescent method using the PIERCE ECL kit (Amersham Biosciences).

Cells were pretreated with 0.5, 1, & 2 μM PA for 1 h, & then stimulated with TGF-beta 1 (5 ng/mL) for 24 h. Total RNA extracted from cells was analyzed by the real-time polymerase chain reaction to determine alpha -SMA & ColIa1 mRNA expression; (C) the total protein extracted from cells was subjected to Western blotting to determine alpha -SMA & collagen I alpha 1 expression. Protein bands were imaged using densitometry & analyzed using ImageJ software. The relative expression levels of target proteins were normalized using beta -actin as an internal control. The vimentin antibody stains the intermediate filament network while the actin antibody labels the submembranous cytoskeleton, stress fibers, and bundles of actin associated with cell adhesion sites.

Western Blot: Collagen I alpha 1 Antibody - Azide and BSA Free [NBP1-30054] - TNC deficiency reduced kidney fibrosis in animal models.Deletion of TNC (TNC−/−) significantly attenuated the induction of collagen I assessed by IF following UUO by approximate 30% at day 7 & 10 compared with their wild-type littermates (A, n = 4 for each time point, two-way ANOVA p < 0.05). Consistently, the expression of fibrosis markers, such as collagen I alpha , fibronectin & plasminogen activator inhibitor-1 (PAI-1), were significantly lower in TNC−/− mice at UUO day 7 & 10 (B, n = 4 for each time point, two-way ANOVA, p < 0.05). Western blot showed that the proteins of collagen I alpha & alpha -SMA were also significantly reduced in TNC−/− mice at UUO day 7 (C, n = 7, p < 0.05). Flow Cytometry: OCT4 Antibody [NB100-2379] - An intracellular stain was performed on Jurkat with NB100-2379 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody.

Immunocytochemistry/Immunofluorescence: OCT4 Antibody [NB100-2379] - Ntera2 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NB100-2379 at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.
