![Western Blot: NOX1 Antibody [NBP1-31546] - Blueberry polyphenol extract (BPE) did not affect the expression of NADPH oxidases (NOX) in angiotensin (Ang) II-treated human aortic endothelial cells (HAECs). HAECs were treated with 200 µg/mL of BPE for 1 h then treated with 200 nM of Ang II for 12 h. Protein expression of NOX1 (A,B), NOX2 (C,D), NOX4 (E,F), & NOX5 (G,H) were determined by Western blot. Quantification was performed using Image Lab (Bio-Rad Laboratories, Inc.). Data are expressed as mean ± SD from three (NOX1 & NOX4) & five (NOX2 & NOX5) independent experiments. Cells were seeded onto cell-tak coated 48 mm coverslips in a 48-well plate. Cells were fixed with 2% formaldehyde for 20 min, washed with PBS. After fixation, cells were blocked with 2.0% BSA in PBS for 1 h at room temperature. All the cells were incubated with a primary antibody for anti-rabbit CXCR4 for 1 h. MDA, PC3, KG1 cells were used for the optimization experiments. Cells were seeded onto cell-tak coated 48 mm coverslips in a 48-well plate. Cells were fixed with 2% formaldehyde for 20 min, washed with PBS. After fixation, cells were blocked with 2.0% BSA in PBS for 1 h at room temperature. All the cells were incubated with a primary antibody for anti-rabbit CXCR4 for 1 h. After washing with PBS, cells were put in respective secondary antibodies-anti-rabbit dylight 405 for 1 h. ![<b>Biological Strategies Validation. </b>Immunohistochemistry-Paraffin: CCL2/MCP1 Antibody [NBP1-07035] - IHC analysis of a formalin-fixed paraffin-embedded (FFPE) human breast carcinoma tissue section using 1:1000 dilution of CCL2/MCP1 antibody (NBP1-07035) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2. HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). Supershift assay showed that the HIF-1/DNA complex was shifted up in the presence of wild-type (wt) oligo probe and 4 ug HIF-1 alpha antiboby (lanes #13 & 14).
