![Immunocytochemistry/Immunofluorescence: Adenosine A2aR Antibody (7F6-G5-A2) [NBP1-39474] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Adenosine A2a R (7F6-G5-A2), NBP1-39474, at a 1:50 dilution overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). ![Western Blot: Melatonin Receptor 1B Antibody - BSA Free [NLS932] - Effects of luzindole on inhibitory action of melatonin on TGF-beta 1-stimulated proliferation & activation in renal interstitial fibroblasts. (A) Reverse transcription-polymerase chain reaction analysis of melatonin receptor type 1A (MT1) & type 1B (MT2) in NRK-49F cells. (B) NRK-49F cells were treated with with TGF-beta 1 (5 ng/mL) after preincubation with vehicle (Veh) or melatonin (Mel; 1mM) in the presence or absence of luzindole (Luz; 100 μM) for 30 min. Cell viability was assessed using CCK-8 assay at 0, 24, & 48 h. The OD was measured at 450 nm. (C) Western blot analysis for alpha -SMA & p-Smad2/3. Bands are identified as glycosylated & nonglycosylated based on reports cited in the text, but could include interaction with other proteins. For each GPER western, the membrane was stripped & reprobed for beta -actin as a loading control. Quantitation of GPER was evaluated by summing all immunoreactive bands & dividing by beta -actin, then normalizing to HBEC2-KT in each blot.
