![Immunohistochemistry-Paraffin: CXCR3 Antibody [NB100-56404] - IHC analysis of a FFPE human spleen using 1:25 dilution of CXCR3 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of HIER using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (40X) was performed using Aperio AT2 (Leica Biosystems). ![Flow Cytometry: Adenosine A1R Antibody [NB300-549] - Analysis of Adenosine Receptor A1 was done on A549 cells. Cells were fixed with 70% ethanol for 10 minutes, permeabilized with 0.25% Triton (R) X-100 for 20 minutes, and blocked with 5% BSA for 30 minutes at room temperature. Cells were labeled with Adenosine Receptor A1 Rabbit Polyclonal Antibody (PA1041A, red histogram) or with rabbit isotype control (yellow histogram) at 3-5 ug/million cells in 2.5% BSA. After incubation at room temperature for 2 hours, the cells were labeled with Alexa Fluor (R) 488 Goat Anti-Rabbit Secondary Antibody (A11008) at a dilution of 1:400 for 30 minutes at room temperature. The representative 10,000 cells were acquired and analyzed for each sample using an Attune (R) Acoustic Focusing Cytometer. ![Immunocytochemistry/Immunofluorescence: LPAR1/LPA1/EDG-2 Antibody [NBP1-03363] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-EDG2/LPAR1/LPA1 (NBP1-03363) at a 1:100 dilution overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin was used as a co-stain at a 1:1000 dilution and detected with and anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). ![Immunohistochemistry-Paraffin: LPAR1/LPA1/EDG-2 Antibody [NBP1-03363] - IHC analysis of formalin fixed paraffin-embedded (FFPE) human prostate cancer using LPAR1 antibody at 1:100 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Staining was obseved in the scattered ducts. In cells transfected with the pcDNA 3.1/c-myc-MasR construct, antibodies NLS-1531 & sc-54682 generated a similar staining pattern to that generated with the anti c-myc antibody. No staining was observed with any of these antibodies in cells transfected with the empty vector pcDNA 3.1. Antibody sc-135063 was capable of staining the plasma membrane of cells overexpressing the MasR but also generated widespread signals in cells transfected with the empty vector. For antibody AAR-013 there was no staining associated with the cell membrane. Intense immunocytochemical staining of identical distribution & intensity was revealed in cells transfected with the empty vector & those transfected with the c-myc-MasR construct. (A) In heart sections, NLS-1531 antibody stained predominantly the cytoplasm of cardiomyocytes with similar intensity in WT & MasR-KO hearts. The AAR-013 antibody stained the cardiomyocytes nucleus & weaker staining was observed in their cytoplasm. The same pattern was found in heart sections of MasR-KO mice. (B) In kidney sections, for NLS-1531 antibody, staining was mostly restricted to cytoplasm of numerous tubules cells with similar intensity in WT & MasR-KO kidneys. Antibody AAR-013 stained most intensely tubules cells nucleus & weaker staining was observed in their cytoplasm. Glomeruli were also stained positively. The same pattern was found in kidney sections of MasR-KO. Preincubation of the AAR-013 antibody with the blocking peptide provided by the vendors eliminated the immunohistochemical nuclear staining both in WT & MasR-KO mice kidney sections.
