BMMs were incubated with M-CSF (30 ng/ml) and RANKL (40 ng/ml) for 40 h, washed thoroughly, and incubated further with LPS (50 ng/ml) in the presence of M-CSF (30 ng/ml) for 48 h. Cells were transfected with 30 nM of miR-29b mimic or con mimic in the presence of M-CSF (30 ng/ml) for 6 h. Total RNA was analyzed by qPCR to quantify the expression of BMF, PUMA, BAK1, BIM, and HRK. Expression levels with con mimic treatment were set at 1. ![Immunocytochemistry/Immunofluorescence: non-muscle Myosin IIA Antibody (2B3) [H00004627-M03] - Bovine ovarian granulosa cells. Anti Myosin IIA immunofluorescent staining (green). DNA in counterstained with DAPI (blue). Control was obtained by omitting the primary antibody. Fixation: 4% PF for 7 minutes at rt. Permeabilization: 0.1% Triton-X in PBS for 7 minutes. Blocking: 20% donkey serum, 1% BSA in PBS, 1 hour at rt. Primary antibody: 1:50 or 1:100 in 1% BSA/PBS over night at 4C. Secondary antibody: donkey anti mouse -Alexa 488 (Invitrogen), 1:500.
