![<b>Genetic Strategies Validation. </b>Western Blot: Bcl-2 Antibody [NB100-56098] - EA reduced MPP+-induced dopaminergic neuronal apoptosis by increasing BDNF (brain-derived neurotrophic factor) expression and further Akt phosphorylation in the rat substantia nigra. Eight days after MPP+ administration, our Western blot results (MAB7566) show that MPP+ treatment reduced tyrosine hydroxylase and Bcl-2 expression in the ipsilateral side of the rat substantia nigra (SN), but not in the contralateral side. EA stimulation (50 Hz) enhanced mature BDNF, tyrosine hydroxylase, and Bcl-2 expression in the MPP+-treated ipsilateral side. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of BCL2 in the cancer cells with relatively intense signal in their nuclear membranes. ![Western Blot: TERT Antibody (2C4) [NB100-317] - IFI16 inhibits c-Myc-stimulated transcription & hTERT expression in HeLa cells.(A) Total protein extracts prepared from HeLa cells infected with control retrovirus (lane 1) or a virus encoding IFI16 protein (lane 2) were subjected to immunoblotting using antibodies specific to the indicated proteins. ![Immunohistochemistry-Paraffin: HIF-1 alpha Antibody [NB100-654] - IHC analysis of a formalin-fixed paraffin-embedded tissue section of human endometrium carcinoma AN3CA cell line based xenograft using rabbit polyclonal HIF-1 alpha antibody NB100-654 at 1:300 dilution. The signal was developed using HRP-labelled secondary antibody and DAB reagent, and the section was further counterstained using hematoxylin. ![Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). The membrane was probed with 2 ug/ml anti-LC3B Antibody in 1% milk, and detected with an anti-rabbit HRP secondary antibody using chemiluminescence. Rictor silencing was induced by cell transfection with rictor siRNA and then, cells were stimulated with IGF-1 for 24 hours in presence or absence of E2. mTORC2 downregulation was confirmed by abolished Akt-pS473 and resulted in decreased SERCA2A protein expression. ![Western Blot: Rictor Antibody [NB100-612] - mTOR complex protein expressions & rictor phosphorylation at Ser1235. Cardiomyocytes were pretreated for 30 min with 20 nM rapamycin, then stimulated with 10 nM IGF-1 in presence/absence of 10 nM E2 for 24 h. Western blots for GSK3beta-pS9, GSK3beta, rictor pS1235 and rictor. At least 3 independent experiments indicate that rictor phosphorylation at S1235 by GSK-3beta downregulates mTORC2 activity. IGF-1 induced strong phosphorylation of GSK-3beta at Ser9 in the absence and presence of E2. Rapamycin pretreatment reduced phosphorylation in E2 co-treated cardiomyocytes, indicating increased activity of GSK-3beta, which was not associated with increased phosphorylation of rictor at S1235.
