Immunocytochemistry/Immunofluorescence: PCNA Antibody (PC10) [NB500-106] - PC12 cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-PCNA Antibody (PC10) NB500-106 at 1 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: PCNA Antibody (PC10) [NB500-106] - HeLa cells were fixed and permeabilized for 10 minutes using cold (-20C) MeOH. The cells were incubated with anti-PCNA [PC10] at 5 ug/mL overnight at 4C and detected with an anti-mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: PCNA Antibody (PC10) [NB500-106] - An intracellular stain was performed on HepG2 cells with PCNA Antibody (PC10) NB500-106G (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 488.

Western Blot: PCNA Antibody (PC10) [NB500-106] - HeLa cell nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with monoclonal anti-PCNA antibody. Proteins were visualized using a goat anti-mouse secondary conjugated to HRP and a chemiluminescence detection system.

Western Blot: PCNA Antibody (PC10) [NB500-106] - Whole cell protein from human HeLa, A431, mouse Neuro2A and rat PC12 cells was separated on a 4-20% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL anti-Histone h3 in block buffer and detected with an anti-mouse HRP secondary antibody using chemiluminescence.

Western Blot: Ubiquitin Antibody (Ubi-1) [NB300-130] - Analysis of HEK293 cell lysates using mouse mAb Ubiquitin, dilution 1:1000 (Green). [1] protein standard (Red), [2] cells maintained in normal medium, [3] cells treated with 10uM of proteasome inhibitor lactacystin (Lc) for 16hrs. Lysed cells were electrophoresed on 4-20% SDS-PAGE and transferred to PVDF membranes. The smear detected above the 200kDa standard represent accumulation of ubiquitinated proteins in proteasome inhibitor-Lc treated cells. The prominent band at 8kDa corresponds to monoubiquitin. Rabbit pAb to HSP60, dilution 1:5000 (Red) was used as a loading control.

Immunohistochemistry-Paraffin: Ubiquitin Antibody (Ubi-1) [NB300-130] - FFPE section of cerebral cortex of an Alzheimer patient processed with Ubiquitin antibody using HRP/DAB (Brown) and also stained with haemotoxylin (Blue). A typical flame shaped tangle is seen in a pyramidal neuron in the center and is surrounded by some dystrophic neurites, also strongly ubiquitin positive. Both are commonly seen in cortical and hippocampal Alzheimer brain sections and are typical for this disease, but are rare or absent in healthy brain.

TLR4 Inhibitor Peptide Set [NBP2-26244] - TLR4/MD-2/CD14/NF-kB/SEAPorter stably transfected cells were plated in 96-well plates at 1x10^5 cells/well. After 16 h, cells were preincubated with various concentrations of inhibitor or control peptides (CP) for 1 h. Cells were then stimulated with 10 ng/ml LPS for 24 h. The secreted alkaline phosphatase (SEAP) was analyzed using the SEAPorter Assay Kit. Inhibitory effect on the LPS-mediated TLR4 activation by the VIPER peptide is shown in panel A, and IC50 of the VIPER peptide is described in panel B.

Immunocytochemistry/Immunofluorescence: ATG9A Antibody [NB110-56893] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. Western Blot: c-Myc Antibody [NB600-336] - Analysis of whole cell lysate (WCL) of Jurkat cells using anti-cMyc antibody (NB600-336) at 1:1000 dilution. HRP conjugated goat anti-rabbit IgG (H+L) cross adsorbed secondary antibody was used with ECL substrate for the detection of c-Myc antibody bound to the blotted protein. This c-Myc antibody detected the c-Myc specific band at its expected position (48-50kDa). The signal below 10 kDa in this blot is potentially the degraded protein and we have not characterized this band.

Our data indicated that upon acquired resistance in HNSCC cells or in non-responder HNSCC tumors, the levels of miR124-3p and miR766-3p go up, which in turn down-regulate its direct target genes: CREBRF and NR3C2, and consequently the expression of downstream targets of CREBRF (ATG5/CREB3) and NR3C2 ( beta -catenin/c-Myc) increased in resistant tumors, which are positively correlated with poor prognosis. Thus, by enhancing the CREBRF-ATG5/CREB3 and NR3C2-beta -catenin/c-Myc axis, miR124-3p and miR766-3p support aggressive HNSCC progression. Depletion of retromer or retromer-associated SNXs decreases EGFR protein levels. Immunohistochemistry-Paraffin: Calreticulin Antibody [NB600-101] - Analysis of a FFPE tissue section of human thyroid gland using 1:50 dilution of Calreticulin antibody. The signal was developed using HRP-DAB based detection method which followed counterstaining of the nuclei with hematoxylin. This representative section shows a strong positivity of Calreticulin in the follicular epithelial cells, wherein the signal was found to be very intense in the perinuclear region of the cells which correlates well with Endoplasmic reticulum localization of this protein. The para-follicular cells, endothelial cells of blood vessels (not the RBCs though) and the loose connective tissue in the section showed a weak cytoplasmic staining. Some staining was observed in the follicles/colloids also which is potentially the secreted form of Calreticulin.

Immunocytochemistry/Immunofluorescence: Calreticulin Antibody [NB600-101] - The Calreticulin antibody NB600-101 was tested in HeLa cells at a 1:250 dilution against DyLight 488 (Green). Alpha-tubulin and nuclei were counterstained against DyLight 550 (Red) and DAPI (Blue), respectively.

Simple Western: Calreticulin Antibody [NB600-101] - Lane view shows a specific band for Calreticulin in 0.5 mg/ml of HeLa lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

Calreticulin was detected in immersion fixed U-2 OS human osteosarcoma cell line using Rabbit anti-Calreticulin Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NB600-101AF647) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 40X objective.
