![Immunocytochemistry/Immunofluorescence: LAMP-1/CD107a Antibody (5E7) [NBP2-52721] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-LAMP (5E7) at a 1:200 dilution overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). ![Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). Cardiomyocytes were pretreated for 30 min with 20 nM rapamycin, then stimulated with 10 nM IGF-1 in presence/absence of 10 nM E2 for 24 h. Western blots for GSK3beta-pS9, GSK3beta, rictor pS1235 and rictor. At least 3 independent experiments indicate that rictor phosphorylation at S1235 by GSK-3beta downregulates mTORC2 activity. IGF-1 induced strong phosphorylation of GSK-3beta at Ser9 in the absence and presence of E2. Rapamycin pretreatment reduced phosphorylation in E2 co-treated cardiomyocytes, indicating increased activity of GSK-3beta, which was not associated with increased phosphorylation of rictor at S1235. Expression of myc tagged Sin1 constructs can be detected only after induction with Doxycycline (Dox). Cells were treated with 100nM of doxycycline (+) for 72 hours & expressed proteins were detected by immunoblot of whole cell lysates with anti-myc (9E10) antibodies. C. & D. Sin1 constructs incorporate into mTORC2 & displace endogenous Sin1. Constructs were induced for 72 hours prior to immune precipitation. Interestingly, PD-L1 is found at similar levels in both normal donor & glioma patient plasma exosomes & these findings were confirmed by western blot. Western Blot analysis also shows that exosomal markers Flotillin-1 & CD63 are found universally in plasma exosomes from normal donors (n = 3), glioblastoma (grade 4) patients (n = 4), & grade 3 glioma patients (n = 3), though CD63 expression may be slightly reduced in glioma patients. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of phosphor-AKT (Ser-473) in the cancer cells as well as the stromal cells.
