Preparation
Method
Novus' biologically active proteins are stringently purified to provide only the safest and most highly effective proteins available. This protein was expressed in E. coli, purified by HPLC, QC tested by SDS-PAGE and Western Blot and validated on appropriate cell lines for bioactivity. Details of Functionality
TNF Receptor I Protein is fully biologically active when compared to standard. The ED50 as determined by its ability to inhibit the TNF-alpha mediated cytotoxicity in the L-929 cells is less than 0.05 ug/ml, corresponding to a specific activity of > 2 x 10^4 IU/mg in the presence of 0.25 ng/mL of rHuTNF-alpha.

Reconstitution Instructions
Recommended to centrifuge prior to opening. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/mL. Apportion stock solutions into working aliquots and store at <-20C.

Notes

This lyophilized preparation is stable at 2-8 degrees C, but should be kept at -20 degrees C for long term storage, preferably desiccated. Upon reconstitution, the preparation is most stable at -20 to -80 degrees C, and can be stored for one week at 2-8 degrees C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degrees C to -80 degrees C. Avoid repeated freeze/thaw cycles.

Background

TNF Receptor I (TNF RI), also named as TNFR-I, TNF R1, TNFRSF1A, p55 and p60, belongs to the tumor necrosis factor receptor superfamily, and it is encoded by the TNFRSF1A gene in human. TNF Receptor I (TNF RI)is a universally expressed protein and is thought to form homotrimers after ligand binding. TNF Receptor I (TNF RI) is one of the major receptors for the TNF-alpha. This receptor can activate the transcription factor NF-kappaB, mediate apoptosis, and function as a regulator of inflammation. Mature human soluble TNF RI shares 68 % ~ 73 % amino acid sequence identity with murine, rat, bovine and porcine soluble TNF RI.
