This antibody was developed against two synthetic peptides corresponding to amino acids 18-32 (LGWKQGDEEEKWAEK) and amino acids 315-330 (SNVNRNSTIENTRRHI) of human SMAD1. GenBank Accession No. AAP36050.1.
In A549, a 50 kDa band is observed. The two synthetic peptide immunogens (LGWKQGDEEEKWAEK) and (SNVNRNSTIENTRRHI) used to make this antibody are 100% conserved between SMAD1/5/8/9. Therefore, the antibody recognizes SMADs 1, 5, 8, and 9. The antibody may cross-react with SMAD2 or SMAD3. The actual SMADs detected with this antibody may vary according to cell or tissue-type specific expression.

MAD-related proteins are a family of intracellular proteins that are essential components in the signaling pathways of the serine/threonine kinase receptors of the transforming growth factor beta superfamily. SMADs can be divided into receptor-regulated SMADs (R-SMADs: SMAD1, 2, 5, 8 and 9), common-mediator SMAD (co-SMAD: SMAD4), and inhibitory SMADs (I-SMADs: SMAD6 and 7). SMAD1, 5, 8 and 9 have high degrees of homology and antibodies are available that recognize sequences common to all of them. SMAD8 and SMAD9 are typically used as alternate names for one another in the literature.
