Human ROR gamma/RORC/NR1F3 Luciferase - (LUCPorter™) Stable Reporter Cell Line Summary

Description
As a constitutively active luciferase reporter cell line, the ROR gamma T Prom/LUCPorter Reporter Cell Line is suitable for screening antagonists/inhibitors that can suppress ROR gamma T induction related with regulatory pathways for Th17 cells. As the nuclear receptor family member of transcription factors, ROR gamma is encoded by a single gene called RORC. ROR gamma is expressed in various tissues including muscle, kidney and liver. A recent study demonstrates that similar to its isoform ROR gamma T, ROR gamma is also preferentially expressed in Th17 cells and is effective in driving Th17 cell differentiation.

This study also shows that ROR gamma and ROR gamma T expression is controlled by two different promoters in Th17 cells. Based on the ROR gamma promoter study (Figure 1), we have developed the luciferase reporter cell line, in which constitutive activation of ROR gamma promoter induces sustained production of the luciferase reporter protein. When tested with several proinflammatory signal inhibitors including Curcumin (NBP2-26243), Celastrol (NBP2-29362), Plumbagin (NBP2-26242), Thymoquinone (NBP2-26241) and Piceatannol, the ROR gamma promoter activity in this cell line was differentially suppressed by those inhibitors (Figure 2).

Notes

The HEK 293 cell line used to generate this reporter line is an immortalized human cell line. Any human lines should be considered as biohazard and be used in a Biosafety II laboratory. Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed. - Use pipette aids to prevent ingestion and keep aerosols down to a minimum. - No eating, drinking or smoking while handling the ROR gamma T reporter cell line. - Wash hands after handling the ROR gamma T reporter cell line and before leaving the lab. - Decontaminate work surface with disinfectant or 70% ethanol before and after working with the cell line. - All waste should be considered hazardous. - Dispose of all liquid waste after each experiment and treat with bleach.

Background

ROR gamma/RORC/NR1F3, aka RAR Orphan Receptor gamma, is a Nuclear Receptor 1 Thyroid Hormone-Like receptor containing a short A-T rich sequence follow by a single core motif half-site 5'-AGGTCA-3'. RORC is a key regulator of thymopoiesis, bone metabolism, T-cell apoptosis, and lymphoid organogenesis. NR1F3 regulates intrinsic transcriptional functions. Unlike other major nuclear receptors, RORC binds as a monomer to hormone response elements and is documented in mouse thymus, adipose, bone, skeletal muscle, liver, kidney, and human skeletal muscle. An N-terminal isoform of ROR gamma, ROR gamma T, inhibits Fas ligand expression and cytokine secretion in immature thymocytes.
