Chromatin Immunoprecipitation: RNA Polymerase II/POLR2A Antibody [NBP2-32080] - ChIP-qPCR analysis of RNA polymerase II binding at pause site of the Jun B gene. Chromatin isolated from one 15 cm plate of HeLa cells was immunoprecipitated with 8.0 ug of anti-RNA polymerase II antibodies and PCR amplified.

Immunoprecipitation: Rabbit Polyclonal RNA Polymerase II/POLR2A Antibody [NBP2-32080] - Detection of human RNA Polymerase II by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-RNA Polymerase II antibody (NBP2-32080 lot 2) used for IP at 6 ug per reaction. RNA Polymerase II was also immunoprecipitated by a previous lot of this antibody and a second antibody against a different epitope of RNA Polymerase II antibody. For blotting immunoprecipitated RNA Polymerase II, the second antibody was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 75 seconds.

Background

RNA polymerase II (RNAPII) is an essential component of the RNAP II transcription elongation complex. Recruitment of RNA processing factors to the elongation complex is coordinated by the phosphorylation of Serine-2 and Serine-5 residues within the C-terminal repeat (YSPTSPS) domain of the largest subunit of RNAPII.

• Comments: ***Bio-Techne Response: This review was submitted through the legacy Novus Innovators Program, reflecting a new species or application tested on a primary antibody.*** Hep3B cells were lysed in immunoprecipitation lysis buffer and incubated 20 min on ice. Following centrifugation at 18,000 x g at 4C, lysates were precleared with protein G-Sepharose for 2 h with rotation. 1 mg of lysate was incubated with rotation overnight at 4C with 2 ug of NBP2-32080 antibody, followed by incubation with protein G-Sepharose for 2 h at 4C and washes with IP lysis buffer 4x for 5 min at 4C. Immunoprecipitates were used for immunoblot assay. WB was performed using a 1:1000 dilution of RNA Pol II antibody in 1% BSA/PBS.
