Background

RMND5A, also known as Protein RMD5 homolog A, consists of a 391 amino acid long isoform that is 44 kDa and a 98 amino acid short isoform that is 11 kDa, and it is involved in protein binding and required for meiotic nuclear division. Studies of RMND5A are being done on the following diseases and disorders: lissencephaly, malaria and ductus arteriosus. This protein has been known to have interactions with SHBG, SMAD9, C20orf11, GID8, POLR3H, YPEL4, RANBP9, MKLN1, KLHL2 and CCT3 proteins.

Western Blot: GLS2 Antibody [NBP1-76544] - Glutaminase protein expression in PC3 and PC3M cell lines before/after CB-839 treatment. PC3 & PC3M cells were treated (+) or not treated (-) with GLS1 inhibitor CB-839 (1 uM) and their expression of GLS1 & GLS2 proteins determined by Western blot. Antibodies specific for GAC & KCA were utilized on two replicate samples, while GLS2 expression was determined on three. PC3 cell lysates expressed GLS2 more than PC3M cell lysates regardless of CB-839 treatment. The difference in GLS2 expression was significant in the untreated cells (P?<?0.03) but because of high variability the difference was not significant in the drug-treated cells. Levels of GLS1 (GAC and KGA) were similar in all cell lysates. Western blot shows lysates of C2C12 mouse myoblast cell line, Jurkat human acute T cell leukemia cell line, K562 human chronic myelogenous leukemia cell line, and NR8383 rat alveolar macrophage cell line. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human EMP/MAEA Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7288) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for EMP/MAEA at approximately 45 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
EMP/MAEA was detected in immersion fixed Jurkat human acute T cell leukemia cell line using Sheep Anti-Human EMP/MAEA Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7288) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue). The cells were incubated with anti-ROR gamma overnight at 4C and detected with an anti-rabbit Dylight 488 (Green). Alpha tubulin (DM1A) NB100-690 was used as a co-stain and detected with an anti-mouse Dylight 550 (Red). Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
