Background
Siglecs (sialic acid-binding Ig-like lectins)
are I-type (Ig-type) lectins belonging to the Ig superfamily (1, 2). They are
characterized by an N-terminal Ig-like V-type domain which mediates sialic acid
binding, followed by varying numbers of Ig-like C2-type domains (3, 4). Siglec 5 to 11 are collectively referred to as
CD33-related Siglecs due to their high degree of sequence similarity with
CD33/Siglec-3 both in their extracellular and intracellular regions. One
remarkable feature of the CD33-related Siglecs is their differential expression
pattern within the hematopoietic system (4, 5). Mouse Siglec-H is an endocytic receptor expressed predominantly on a specialized subset of
leukocytes, the plasmacytoid dendritic cell (pDC). Siglec-H
has not been shown to bind sialic acids (6). Unlike
other CD33-related siglecs, the cytoplasmic tail of Siglec-H lacks ITIM-related
sequence (6, 7). The
transmembrane region contains a charged lysine residue that could associate
with DAP12 which reduces interferon-alpha (IFN alpha ) production (6, 8).
