Specificity
It recognizes a glycoprotein of 75kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). Its epitope spans in aa 1-160 of extracellular domain of NGFR/NTR. NGF-receptor contains an extracellular domain containing four 40-amino acid repeats with 6 cysteine residues at conserved positions followed by a serine/threonine-rich region, a single transmembrane domain, and a 155-amino acid cytoplasmic domain. The cysteine-rich region contains the nerve growth factor binding domain. NGF is important for the development, differentiation, and survival of variety of neuronal and non-neuronal cells. Its action is mediated by binding two distinct receptors, the high affinity p140 and low affinity p75.

Western blot shows lysates of RPMI 8226 human multiple myeloma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human TrkA Antigen Affinity-purified Polyclonal Antibody (Catalog # AF175) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for TrkA at approximately 140 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
TrkA was detected in immersion fixed paraffin-embedded sections of human colon using Goat Anti-Human TrkA Antigen Affinity-purified Polyclonal Antibody (Catalog # AF175) at 3 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit  (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Recombinant Human beta ‑NGF (Catalog # 256-GF) stimulates proliferation in the TF‑1 human erythroleukemic cell line in a dose-dependent manner (orange line). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
LEDGF was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human LEDGF Monoclonal Antibody (Catalog # MAB3468) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red, upper panel; Catalog # NL007) and counterstained with DAPI (blue, lower panel).
