Western Blot: Neprilysin/CD10 Antibody (FR4D11) - Chimeric [NBP3-12068] - Daudi cell lysate (35ug protein in RIPA buffer) was resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of FR4D11 (NBP3-12068) at 2 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Immunocytochemistry /Immunofluorescence: Neprilysin/CD10 Antibody (FR4D11) - Chimeric - Azide and BSA Free [NBP3-12068] - Immunofluorescence analysis of paraformaldehyde fixed Daudi cells on Shi-fix(TM) coverslips stained with NBP3-12068 at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-12068, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Background

CD10, also known as the Common Acute Lymphocytic Leukemia Antigen (CALLA), is a cell surface enzyme with neutral metalloendopeptidase activity which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, immature B cells with in adult bone marrow and on cells from patients with chronic myelocytic leukemia (CML). CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, with especially high expression on the brush border of kidney and gut epithelial cells.
