Background
FUNCTION: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity. MPO is an important marker for myeloid cells, from the promyelocyte stage and to the mature forms. CATALYTIC ACTIVITY: Donor + H2O2 = oxidized donor + 2 H2O. CATALYTIC ACTIVITY: Cl- + H2O2 = HOCl + 2 H2O. COFACTOR: Binds 1 calcium ion per heterodimer. COFACTOR: Binds 1 heme B (iron-protoporphyrin IX) group covalently per heterodimer. SUBUNIT: Tetramer of two light chains and two heavy chains. SUBCELLULAR LOCATION: Lysosome. ALTERNATIVE PRODUCTS: 3 named isoforms produced by alternative splicing. DISEASE: Defects in MPO are the cause of myeloperoxidase deficiency (MPD). MPD is an autosomal recessive defect that results in disseminated candidiasis. SIMILARITY: Belongs to the peroxidase family. XPO subfamily.

Our laboratory would like to assess the extent of leukocyte infiltration into rabbit kidney induced by metabolic acidosis. We are currently exploring the feasibility of a myeloperoxidase assay utilizing antibody capture of myeloperoxidase for this purpose. Rabbit myeloperoxidase shares 88% and 86% identity with the human and mouse orthologues, respectively. Can you determine based on the immunogen, antibody epitope, or from direct testing data whether this antibody will cross-react with the rabbit orthologue?
- Unfortunately, we have not mapped the actual epitope nor have we tested directly in rabbit. Based on the immunogen pf 646-743 aa. homology to rabbit we would say that there is definite cross reactivity with 85% homology. VSEP LKRKGRVGPL LACIIGTQFR KLRDGDRFWW ENEGVFSMQQ RQALAQISLP RIICDNTGIT TVSKNNIFMS NSYPRDFVNC STLPALNLAS WREAS I did check the other MPO antibodies and the problem is we do not have the exact epitope for any and when they use the full length for immunization then we cannot say for certain. However, with full length homology being 88% from human to rabbit is no good region of exclusivity.
