A biologically active recombinant protein with a 16 residue His tag and corresponding to 159 total residues of IFNG.

Source:E. coli The recombinant protein migrates as two bands with an apparent molecular mass of 25 and 27 kDa in SDS-PAGE.

Produced in the endosperm tissue of barley grain (Hordeum vulgare), that exhibits up to 50 times less protease activity than E.coli or mammalian cells. Barley seed is void of any human or animal viral contaminants that could jeopardize your cell culture.
Details of Functionality
The bioactivity of recombinant human IFN-gamma was determined by its dose dependent cytostatic effect, in the presence of TNF-alpha, on HT-29 (human carcinoma) cells. The ED50 value for this batch of IFN-g is 0.12 ng/ml, corresponding to 8.5 x 10e6 U/mg specific activity.

Background

IFN-gamma, a type II interferon and an acid-labile interferon, is mainly produced by CD4 and CD8 T-lymphocytes and natural killer cells activated by antigens, mitogens or alloantigens. IFN-gamma shows no significant homology with IFN-beta or IFN-alpha. IFN-gamma has antiviral and antiparasitic activities and exhibits also immunoregulatory and proinflammatory activities. IFN-gamma induces cytokine production and promotes antigen presentation by up regulating the expression of MHC proteins as well as other antigen proteins. In contrast to other interferons, which are mostly antiviral, the main activity of IFN-gamma seems to be immunomodulatory. Human IFN-gamma is species-specific and is only biologically active in human and primate cells.

  1. In human myeloid cell culture, for 1ml of medium with 2x10^6 cells, how much IFN gamma do you recommend to use, in order to simulate cancer's level?
    • Our interferon gamma protein with catalogue number NBC1-21339 has been validated for functional assays, and the bio-activity was determined by its dose dependent cytostatic effect, in the presence of TNF-alpha, on HT-29 cells. The ED50 value for the current batch of IFN-g is 0.12 ng/ml, corresponding to 8.5 x 10e6 U/mg specific activity. Since you are using a different cell line and a different system, you will need to optimise the assay conditions yourself. I would suggest that you titrate the protein, and try a number of different time points, in order to identify the relevant conditions for your experiment. I am sorry not to be able to provide more detailed recommendations but, as you know, every experimental system is different and requires its own optimisation.
