Background
HAP1 (Huntingtin-associated protein 1) is a cytoplasmic protein that interacts with several proteins such as Huntingtin, p150Glued, KLC, TrkA, TATA-binding protein, proBDNF etc. and plays a key role in microtubule-dependent transport, vesicular trafficking, nerve growth, transcriptional regulation and signal transduction. HAP1 was originally discovered as huntingtin interacting neuronal protein (htt), the Huntington's disease protein, however, unlike htt (expressed ubiquitously throughout body), HAP1 is enriched in brain with abundant expression in the hypothalamus, amygdala, brain stem etc. HAP1 has been associated with microtubule-dependent transporters dynactin p150 and kinesin light chain, as well as with transport of various vesicular proteins or membrane receptors, including BDNF-containing vesicles, EGFR, type 1 inositol (1,4,5)-triphosphate receptor, gamma -aminobutyric acid, type A, receptor, and TRKA. Hap1 is also implicated in microtubule-dependent trafficking in neurons, vesicular transport, gene transcription regulation, membrane receptor trafficking and other functions such as calcium release and protein aggregation.
