Immunohistochemistry-Paraffin: Granzyme B Antibody [NB100-684] - Formalin fixed paraffin embedded human tonsil stained with Granzyme B antibody
Tested applications:* Immunohistochemistry (Formalin-fixed paraffin-embedded sections) - 1/50 - 1/100 for 30mins at RT (ABC method). Perform heat mediated antigen retrieval (using 10mM citrate buffer pH 6.0) before commencing with IHC staining protocol. Immunohistochemistry (Frozen sections) - 1/50 - 1/100 for 30mins at RT (ABC method). Fix with ice cold acetone. Immunofluorescence - 1/50 Not tested in other applications. *Optimal dilutions / concentrations should be determined by the end user. Immunohistochemistry: use at a dilution of 1/50 - 1/100 for 30mins at RT (ABC method). For formalin fixed paraffin embedded sections, perform heat mediated antigen retrieval using 10mM citrate buffer pH 6.0 before commencing with the IHC staining protocol.

The lytic enzymes of cytotoxic lymphocytes seem to be contained within lysosome-like granules found in the cytoplasm. These granules contain perforin, a membrane pore-forming protein, and a family of serine proteases (granzymes). Granzyme B, a 32 kDa protein, is a serine Aspase associated with the granules of Natural Killer (NK) cells. Granzyme B seems to be able to elicit cell death, perhaps by activating an endogenous pathway of programmed cell death.

Caspase 3, the executioner of apoptosis Caspase-3 enzyme is a member of the family of endoproteases which regulate inflammation and apoptosis signaling networks. Caspase-3 is known as an executioner caspase in apoptosis because of its role in coordinating the destruction of cellular stru... Read full blog post.
Caspase 9: The Suicidal Cell Whisperer Cell death via apoptosis is a key cellular function triggered by the cell death receptor family and their ligands which signal through downstream adaptor molecules and the caspase protease family. Among the subclass of initiator caspases that include... Read full blog post.
Caspase 7: The Cell's Suicide Switch Caspase 7 (also known as CASP7, Mch3, ICE-LAP3, CMH-1) is a member of caspase family of cysteine proteases. It is an apoptosis-related cystein peptidase encoded by the CASP7 gene in humans. CASP7 homologous sequences have been identified in nearly all... Read full blog post.
