Recombinant Human Granulin Protein [NBP1-99540] - Chemotaxis effect of Progranulin in THP-1 Cells: Cell migration of THP-1monocytic leukemia cells was evaluated in disposable 24 well transwellpolystyrene membrane with 8um size pores. 0ng/ml, 10ng/ml, 100ng/ml and250ng/ml of Progranulin were diluted in RPMI 1640 supplemented with 0.5%FBS respectively. In case boiled Human Granulin Protein, proteins were boiled at 100 degrees C for 10min. And then Human Granulin Protein samples were added to the lower chamber. THP-1 cells (2 X 105 cells per well), in the same media with Progranulin, were added to the upper chamber. After 16 hours incubation at 37 degrees C in 5% CO2 humidified atmosphere, migration cells in the lower chamber were counted. Migrated cells in two separate fields per well from triplicate wells were enumerated on a hemacytometer by means light microscopy.

Background

Progranulin, also called proepithelin and PC cell-derived growth factor, is a single precursor protein of granulins which are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. Granulins are a variety of active, 6.0 kDa peptides and named granulin A (epithelin 1), granulin B (epithelin 2), granulin C, etc. Both the peptides and intact progranulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis. Progranulin Human Recombinant fused to FLAG at C-terminus produced in HEK is a single, glycosylated, polypeptide chain containing 593 amino acids and having a molecular mass of 88.0 kDa. The Progranulin is purified by standard chromatographic techniques.
