Specificity
Recognizes a protein of 94kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Grp94 has been briefly studied as a prognostic factor in breast cancer.

Hu-chromosome location: 12q23.3 Immunohistochemistry-Paraffin 0.5 - 1.0 ug/ml for 30 minutes at RT; Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10 - 20 min followed by cooling at RT for 20 minutes. The 7mL size is a pre-diluted size and no additional dilutions are required before using this item for the intended application.

Western Blot: GRP78/HSPA5 Antibody [NBP1-06274] - Total protein from human HeLa and A431 cells, mouse 3T3 cells and rat PC12 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL anti-GPR78 in blocking buffer and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: GRP78/HSPA5 Antibody [NBP1-06274] - NIH-3T3 cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-GRP78/HSPA5 at 2 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on NIH3T3 cells with GRP78/HSPA5 Antibody NBP1-06274AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Immunohistochemistry-Paraffin: Calreticulin Antibody [NB600-101] - Analysis of a FFPE tissue section of human thyroid gland using 1:50 dilution of Calreticulin antibody. The signal was developed using HRP-DAB based detection method which followed counterstaining of the nuclei with hematoxylin. This representative section shows a strong positivity of Calreticulin in the follicular epithelial cells, wherein the signal was found to be very intense in the perinuclear region of the cells which correlates well with Endoplasmic reticulum localization of this protein. The para-follicular cells, endothelial cells of blood vessels (not the RBCs though) and the loose connective tissue in the section showed a weak cytoplasmic staining. Some staining was observed in the follicles/colloids also which is potentially the secreted form of Calreticulin.

The membrane was probed with 5 ug/ml anti-ATF6 and detected with an anti-mouse HRP secondary antibody using a maximum sensitivity ECL reagent. Note the loss of the 90 kDa modified ATF6 protein (arrowhead) upon DTT treatment to activate the unfolded protein response.
