Application Notes
This Factor XII/F12 antibody is useful for Western Blot, Immunocytochemistry/Immunofluorescence, and IHC-paraffin embedded sections. In Western Blot, bands are seen ~67, 50, and 140kDa representing the full product, a clevage chain product, and a possible dimer. In ICC/IF, cytoplasmic staining was observed in NIH/3T3 cells. In IHC-P, staining was observed in the cytoplasm and nucleus of human renal cancer tissue. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

Background

Coagulation factor XII (F12, Hageman factor, HAF, HAE3, HAEX), is a serum glycoprotein that is part of the peptidase S1 family. It is involved in hemostasis, blood coagulation, fibrinolysis, and the production of bradykinin and angiotensin. F12 is a secreted protein that can serve as a hydrolase, protease, and serine protease. F12 is triggered by kallikrein in alpha-factor XIIa, which is then further altered by trypsin into beta-factor XIIa. F12 has one known isoform, and the protein contains two EGF-like domains, a fibronectin type-I domain, a fibronectin type-II domain, a Kringle domain, and a peptidase S1 domain.

Western Blot: Plasminogen Antibody [NB600-930] - Detection of Plasminogen under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified target proteins contained 4% BME and were boiled for 5 minutes. Samples of 1ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:3000 dilution of primary antibody. Detection shown was using Dylight 649 conjugated Donkey anti goat 1 hr RT.

Immunocytochemistry/Immunofluorescence: Fibronectin Antibody [NBP1-91258] - NIH-3T3 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Fibronectin at 2 ug/mL overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at 1:500. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Recombinant Human Granzyme A protein was serially diluted 2-fold and captured by Mouse Anti-Human Granzyme A Monoclonal Antibody (Catalog # MAB29052) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-Human Granzyme A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2905) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).
Western blot shows lysates of NK‑92 human natural killer lymphoma cell line. PVDF membrane was probed with 0.25 µg/mL of Goat Anti-Human Granzyme A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2905) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for Granzyme A at approximately 28 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
