Immunocytochemistry/Immunofluorescence: Ezrin Antibody [NBP2-52977] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with Ezrin Antibody (NBP2-52977) at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: Ezrin Antibody [NBP2-52977] - Total protein from HeLa, Ntera2 and 3T3 cells was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/ml anti-EZR in 1% milk, and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Immunohistochemistry-Paraffin: Ezrin Antibody [NBP2-52977] - IHC analysis of a formalin fixed paraffin embedded mouse kidney tissue section using rabbit anti-Ezrin antibody at 1:50 dilution. The sections were stained using HRP-DAB based detection followed by hematoxylin counterstaining. The antibody generated a membrane-cytoplasmic signal in the epithelial cells of various tubules and glomeruli did not show much of the expression.

Cells were imaged using a 40X objective.

Ezrin was detected in immersion fixed A431 human skin carcinoma cell line using Rabbit anti-Ezrin Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP2-52977AF647) (light blue) at 5 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Background

Ezrin, Moesin and Radixin belong to a family of highly homologous Actin associated proteins that are localized just beneath the plasma membrane. The proteins are believed to be involved in the mediation of interactions between cytoskeletal and membrane proteins. Ezrin serves as a major cytoplasmic substrate of various protein-tyrosine kinases, including the epidermal growth factor receptor. Ezrin has also been identified as a cAMP-dependent protein kinase (A-kinase) anchoring protein and designated AKAP78. Moesin and Radixin share over 70% homology with Ezrin and are coexpressed within various cell types. Despite the high degree of homology, the three proteins exhibit a distinct receptor-specific pattern of phosphorylation.
