The monoclonal antibody E9 reacts with Endoglin a 190 kDa homodimeric transmembrane glycoprotein composed of disulfide-linked subunits. The external domain binds TGF-beta1 and -beta3 isoforms with high affinity. Two different isoforms (L and S) of CD105 with capacity to bind TGF-beta have been characterized which differ in the amino acid composition of their cytoplasmic tails. Mutations in the gene encoding endoglin have been linked to the human disease hereditary hemorrhagic telangiectasia type 1 (HHT1) a vascular disorder characterized by localized vascular dysplasia and a tendency towards arteriovenous malformations. Mice expressing a single CD105 allele develop external signs of disease similar to human HHT1, supporting the haploinsufficiency model for HHT1. Mice lacking endoglin die from defective angiogenesis characterized by failure of vascular smooth muscle investment of embryonic blood vessels, suggesting a defect in vascular smooth muscle cell development. Microvessel density (MVD) has been reported to be an independent prognostic indicator of outcome in a variety of human malignancies, with increased MVD correlating with shorter overall and relapse-free survival rates. The MVD counts using anti-CD105 antibody significantly correlated with overall and disease-free survival. Anti-CD105 monoclonal antibody E9 and anti-CD34 monoclonal antibody have been successfully used to quantify MVD in human breast carcinoma |

The monoclonal antibody E9 can be used for immunohistology on frozen sections, immuno assays as coating and detection, immuno precipitation, Western blotting and flow cytometry. Antibody E9 cannot be used for immunohistology on paraffin sections and forFor immunohistology, flow cytometry and Western blotting dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:10. |

Background
CD105 (Endoglin) is a homodimeric transmembrane glycoprotein serving in presence of TGFAR-2 as a receptor for TGFA-1 and TGFA-3. CD105 is highly expressed on endothelial cells and promotes angiogenesis during wound healing, infarcts and in a wide range of tumours and its gene expression is stimulated by hypoxia. CD105 prevents apoptosis in hypoxic endothelial cells and also antagonises the inhibitory effects of TGFA-1 on vascular endothelial cell growth and migration. Normal cellular levels of CD105 are required for formation of new blood vessels.

I wonder if you have a CD105 or CD34 antibody suitable for IHC that is specific for human and do not bind mouse?
- We do not have any anti-human CD34 or CD105 antibodies that are confirmed to NOT detect the mouse protein. When we have tested an antibody and confirmed that it will not react with mouse samples, we will add Mu(-) to the datasheet, and unfortunately all of our CD105 and CD34 antibodies will either detect the mouse protein, or they have not been used in mouse samples before.

One of our customers wants to do WB with NBP1-42383 to detect CD105 from Rat sample, but before the experiment, he wants to confirm whether this antibody is suitable for detecting CD105 from Rat sample? That is to say, do you have any in-house test for Rat sample?
- Unfortunately this one has the species of rat reported to us by a customer. Therefore we do not have any in house testing data to provide showing reactivity in rat on file. We will still guarantee this species for your customer should they have problems testing in Rat samples and report the difficulty to us within 6 months from purchase.
