Western blot shows lysates of Raji human Burkitt's lymphoma cell line (negative control), HepG2 human hepatocellular carcinoma cell line, and COLO 205 human colorectal adenocarcinoma cell line. PVDF membrane was probed with 1 µg/mL of Rat Anti-Human Angiopoietin‑like Protein 3/ANGPTL3 Monoclonal Antibody (Catalog # MAB38292) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Angiopoietin‑like Protein 3/ANGPTL3 at approximately 51 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background

Angiopoietin-like 3 (ANGPTL3) is a secreted glycoprotein that belongs to the angiopoietin family. It is expressed in the liver and circulates as 70 kDa, 50 kDa, and 32 kDa species. These include full length ANGPTL3 as well as the proteolytically separated N- and C-terminal fragments which contain a coiled coil domain and fibrinogen-like domain, respectively. ANGPTL3 promotes an increase in circulating triglyceride levels by directly inhibiting lipoprotein lipase. Mature human ANGPTL3 shares 77% amino acid sequence identity with mouse ANGPTL3.
