Immunocytochemistry/Immunofluorescence: PML Protein Antibody - BSA Free [NB100-59787] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with PML Protein Antibody (NB100-59787) at 2ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: PML Protein Antibody - BSA Free [NB100-59787] - A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with PML Protein Antibody (NB100-59787) at 1ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunoprecipitation: CRISPR-Cas9 Antibody (7A9-3A3) - N-Terminus [NBP2-36440] - HEK293T expressing N-terminally Flag-tagged S.pyogenes Cas9 were lysed 72h post transfection by resuspending the cells in Hunt buffer and subjecting to 3 freeze-thaw cycles in liquid nitrogen/ice. Proteins were immunoprecipitated from 100ug of whole cell lysate for 1h at 4C with Cas9 supernatant followed by incubation for 1h at 4C with a 1:1 mixture of protein A/G sepharose beads, or for 2h at 4C with Cas9 ab crosslinked to a 1:1 mixture of protein A/G sepharose beads. Beads were washed 2x with Hunt buffer and 1x with TBS. Bound proteins were eluted by boiling in Laemmli, separated by SDS-PAGE and transferred to nitrocellulose. Membrane was blocked, incubated with Cas9 ab, incubated with HRP anti-mouse secondary. *IgG heavy chain

The signal was developed using HRP-labelled anti-mouse secondary antibody and ECL based detection. Observed molecular weight is ~158 kDa.

Immunocytochemistry/Immunofluorescence: CD31/PECAM-1 Antibody (MEC 7.46) [NB100-1642] - MS1 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with at 5.0 ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry-Frozen: CD31/PECAM-1 Antibody (MEC 7.46) [NB100-1642] - Frozen mouse lung tissues of WT B6 and IL1R--/- (knockout) mice were blocked with 1% BSA in PBS and were double stained with Alex647-conjugated CD31/PECAM-1 antibody (MEC 7.46) and PE-conjugated F4/80 for 2 hours at the room temperature. Immunohistochemistry-Paraffin: Collagen IV Antibody [NB120-6586] - Collagen IV antibody at 1:400, 45 min RT showed strong staining in FFPE sections of human kidney (left) with strong red staining observed in glomeruli and liver (right) with strong staining in sinusoids. Staining for both tissues was consistent with a basement membrane distribution. Slides were steamed in 0.01 M sodium citrate buffer, pH 6.0 at 100C for 20 minutes for antigen retrieval.

Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 Antibody [PAb 240] NB200-103 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-p53 (PAb 240) [NB200-103] at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-p53 Antibody (PAb 240) NB200-103 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Mouse primary kidney cells treated with 150 μM CoCl2 for overnight were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. HIF‑1 alpha /DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human/Mouse/Rat HIF‑1 alpha /HIF1A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1935) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunohistochemistry-Paraffin: SR-BI Antibody [NB400-113] - Analysis of a FFPE section of human adrenal gland tissue using 1:100 dilution of SR-BI antibody. The staining was developed using HRP-DAB based detection method and the nuclei of the cells were counterstained with hematoxylin. This antibody generated a specific staining of SR-BI/SCARB1 in the glandular cells. The staining was membrane-cytoplasmic in the zona glomerulosa cells while the signal was primarily localized to the membranes of the cells in the zona fasciculata and zona reticularis layers of the adrenal cortex.
