Human umbilical cord endothelial cells (HUVECs) were cultured for 6 hours in the presence of 25 ng/mL of rhTNF-alpha (210-TA, filled histogram) or rested (open histogram) and stained with Mouse anti-human E-selectin/CD62E (BBA16) followed by Goat anti-Mouse APC-conjugated secondary antibody (F0101B). Gates were set based on isotype control (MAB002, data not shown). Staining was performed using our Staining Membrane-associated Proteins protocol.

E-Selectin/CD62E was detected in immersion fixed HUVEC human umbilical vein endothelial cells activated with TNF-a (Catalog # 210-TA-010) using Mouse Anti-Human E-Selectin/ CD62E Monoclonal Antibody (Catalog # BBA16) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). E-Selectin/CD62E was detected in immersion fixed paraffin-embedded sections of human brain (occipital cortex) using 10 µg/mL Mouse Anti-Human E-Selectin/CD62E Monoclonal Antibody (Catalog # BBA16) overnight at 4 °C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained with the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Immunohistochemistry-Paraffin: OLLAS Epitope Tag Antibody (L2) [NBP1-06713] - Different intracellular and transmembrane domains (from Nrx, nSyb, and sdc) were fused to CD19 and expressed into ORNs targeting the DA1 glomerulus using the CD19::Nrx (top), nSyb::CD19 (middle) and CD19::Sdc (bottom), with the R17H02 driver (in green). The brain samples were co-immunostained with antibodies against the OLLAS tag (present in ligand) & against the pre-synaptic protein, BRP (in magenta). The ligand proteins are co-localized with or adjacent to BRP (arrows), demonstrating that all ligands are enriched at the presynaptic terminals of the ORNs; CD19::Nrx was also expressed at strong levels in the axon shaft outside of the glomerulus (arrowheads in top panels). <b>Biological Strategies Validation. </b>Immunohistochemistry: ASC/TMS1 Antibody [NBP1-78977] - Rats were infected by S. pneumoniae or given PBS sham control. 21.5 umoles/kg Mito-Vit-E (MVE) or vehicle was administered orally 30 minutes post-inoculation, and heart tissues were harvested 24 hours later. Heart sections were co-stained with anti-ASC (brown) and haematoxylin (blue). Negative control was stained with secondary antibody alone. Western Blot: ASC/TMS1 Antibody [NBP1-78977] - Whole cell protein from (1) THP1 and (2) HL60 was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 4.0 ug/ml anti-ACS/TMS1 in 1% milk, and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Flow Cytometry: ASC/TMS1 Antibody - BSA Free [NBP1-78977] - An intracellular stain was performed on MCF7 cells with ASC/TMS1 NBP1-78977 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: ASC/TMS1 Antibody [NBP1-78977] - Tested in MCF-7 cells with FITC (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red).

NQO-1 (A180) was detected in immersion fixed U-2 OS human osteosarcoma cell line using Mouse anti-NQO-1 (A180) Protein G Purified Monoclonal Antibody conjugated to DyLight 650 (Catalog # NB200-209C) (light blue) at 10 µg/mL overnight at 4C. Flow Cytometry: PCNA Antibody (PC10) [NB500-106] - An intracellular stain was performed on HepG2 cells with PCNA Antibody (PC10) NB500-106G (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 488.

Adjacent tissue section was processed for immunohistochemistry using mouse anti-human (Novus Biologicals catalog # NB300-164) at 0.3ug/mL with overnight incubation at 4 degrees Celsius followed by incubation with anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). IHC signal is confined to cytoplasm.

Luciferase activity was then directly analyzed using the one-step luciferase reporter assay reagent. Immunohistochemistry-Paraffin: EGLN3/PHD3 Antibody [NB100-303] - LDL Receptor was detected in immersion fixed paraffin-embedded sections of human liver cancer using rabbit anti-human antibody (NB100-303) at 1:3000 overnight at 4C. Tissue was stained using the VisuCyte anti-rabbit HRP polymer detection reagent (VC003) with DAB chromogen (brown) and counterstained with hematoxylin (blue).

Immunocytochemistry/Immunofluorescence: EGLN3/PHD3 Antibody [NB100-303] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-HIF Prolyl Hydroxylase 3 (NB100-3030) at 1:200 overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at 1:500. Alpha tubulin, DM1A (NB100-690) was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry: EGLN3/PHD3 Antibody [NB100-303] - PHD3 was detected in immersion fixed paraffin-embedded sections of human heart using Rabbit Anti-Human EGLN3 polyclonal Antibody (NB100-303) at 5 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte(TM) HRP Polymer Antibody (VC003). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to nuclei.
