Immunocytochemistry/Immunofluorescence: CD31/PECAM-1 Antibody [NBP1-71663] - HUVEC cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with CD31/PECAM-1 Antibody at 5 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: CD31/PECAM-1 Antibody [NBP1-71663] - CD31/PECAM-1 Antibody was tested in WeHi cells with Dylight 488 (green) alongside a matched isotype control (black).

Western Blot: CD63 Antibody (MEM-259) - BSA Free [NB100-77913] - Cytokine & checkpoint molecule arrays from plasma exosomes showed a decreased concentration of IFN-gamma , IL-10, IL-13, B7-1, B7-2, & ICOSL in GBM patients in comparison to normal donors. (A) Quantification of IFN-gamma , IL-10, & IL-13 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group, *P < 0.05). (B) Quantification of B7-1, B7-2, & ICOSL in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). (C) Quantification of the immunosuppressive checkpoint & T cell costimulatory homolog protein PD-L1 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). Interestingly, PD-L1 is found at similar levels in both normal donor & glioma patient plasma exosomes & these findings were confirmed by western blot. Western Blot analysis also shows that exosomal markers Flotillin-1 & CD63 are found universally in plasma exosomes from normal donors (n = 3), glioblastoma (grade 4) patients (n = 4), & grade 3 glioma patients (n = 3), though CD63 expression may be slightly reduced in glioma patients. Dual RNAscope ISH-IHC: CD14 Antibody (M5E2) [NB100-77758] - TNFA mRNA (red) and CD14 protein (green) were detected in formalin-fixed paraffin-embedded tissue sections of human malignant lymph node. ACD's Integrated Co-Detection Workflow was performed using ACD RNAScope Probe Hs-TNFA and CD14 antibody (M5E2) at 1:200 dilution. Tissue was stained on Leica Bond RX using RNAscope (TM) 2.5 LS Reagent Kit-RED, BOND Polymer Refine Detection (DAB) and Hematoxylin, BOND Polymer Refine Red Detection and Hematoxylin and RNAscope (TM) 2.5 LS Green Accessory Pack. Tissue was counterstained with 50% hematoxylin (blue).

Immunohistochemistry-Paraffin: TfR (Transferrin R) Antibody (OX26) [NB200-585] - Analysis of FFPE mouse spleen using TfR (OX26) antibody at 1:50 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Plasma membrane staining was observed. Staining was performed by Histowiz.

Flow Cytometry: TfR (Transferrin R) Antibody (OX26) [NB200-585] - An intracellular stain was performed on PC12 cells with TfR (Transferrin R) [OX26] Antibody NB200-585AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Analysis of a FFPE tissue section of human adipose using 1:200 dilution of CD36 antibody (NB400-145). The staining was developed using HRP labeled anti-rabbit secondary antibody and DAB reagent, and nuclei of cells were counter-stained with hematoxylin.
