Immunohistochemistry-Paraffin: beta-Defensin 3 Antibody [NB200-117] - Distribution of hBD-2 and hBD-3 in normal oral epithelium. Formalin fixed and parafilm embedded normal human oral tissue was labeled with an anti-hBD-2 specific primary antibody, and detected with an AF488 conjugated secondary antibody. hBD-3 was labeled with a rabbit anti-hBD-3 (Novus Biologicals Inc., Littleton, CO, USA), specific primary antibody and detected using an AF594 (purple) conjugated donkey anti-rabbit (Invitrogen). Note green fluorescence detection of hBD-2 localized to the stratum spinosum and stratum granulosum, while pink/red fluorescence detects hBD-3 exclusively to the stratum basale. hBD-1, not shown, is localized to the same regions as hBD-2. Background

A novel, nonhemolytic antimicrobial peptide (human beta-defensin-3, hBD-3) was isolated from human lesional psoriatic scales and clones from keratinocytes. hBD-3 demonstrated a salt-insensitive broad spectrum of potent antimicrobial activity against many potentially pathogenic microbes including multiresistant S. aureus and vancomyosin-resistant Entoerococcus faecium. Keratinocytes and airway epithelial cells are cellular sources of hBD-3. Tumor necrosis factor alpha and contact with bacteria were found to induce hBD-3 mRNA expression. hBD-3, therefore, might be important in the innate epithelial defense of infections by various microorganisms seen in skin and lung, such as cystic fibrosis.

  beta-Defensin 3 antibody (affinity-purified rabbit anti-human hBD-3 IgG) used for IHC-P on tissues that were collected from the inner side of the pseudocapsule and/or synovial membrane-like interface membrane during operations performed for septic hip or knee implant loosenings in human patients. The antibody was used at 0.25 ug/ml concentration for autostaining the sections on Leica-Bond-Max immunostainer with Bond Polymer Refine HRP labeled polymer -DAB development method. The antibody also used for double-immunofluorescence staining in IHC-P which involved - HIER using sodium citrate pH 6 with microwave, permeabilization with Triton X-100 for 10 min, blocking with 10 % normal goat serum, incubation of sections with 4ug/ml primary antibody for ON at 4C, detection with Alexa conjugated secondary antibodies.

1. Are there by chance any WB images available for NB200-117 that haven’t been posted yet? If not, do you happen to know what band size(s) were actually detected? We have a customer who is concerned because he has encountered several products that are detecting around 15-17 kDa as opposed to the expected mw of ~8 kDa.
   • We unfortunately do not have any images available for this product in western blot. I am sorry for any inconvenience. Since western blot is listed on our datasheet, we will of course fully guarantee the application. Typically a band is seen around 10kDa. Depending on the molecular weight marker, I would expect that there might be some difference seen at such a low molecular weight. 15-17kDa does seem a bit too high. I noticed that the protein does have several disulfide binding points. I would make sure your customer is properly boiling their samples (we recommend 10 minutes at 95C) and that they are adding at least 5% reducing agent to their sample buffer. We use BME typically, but DTT is also good.
