Western Blot: NLRP3/NALP3 Antibody - BSA Free [NBP2-12446] - Exercise training prevents HFD-induced upregulation of cardiac pyroptosis. Western blotting was performed to determine the components of inflammasome complex & pyroptosis in the left ventricle of mice hearts. (A) HFD-fed obese mice show upregulation of the NLRP3 inflammasome in the left ventricle, which is prevented by exercise training. (B) Apoptosis-associated speck adaptor protein (ASC) that assembles with NLRP3 & caspase-1 to make inflammasome complex is elevated in the heart of obese mice but attenuated by exercise training. (C) Exercise training prevents HFD-induced upregulation of cardiac caspase-1. (D) Expression of IL-1 beta , which is upregulated by HFD, is normalized by exercise training. The C57BL/6 mice were subconjunctivally injected with 6 μL (4 × 108 PFU) of the Ad-GFP-shRNA or Ad-NLRP3-shRNA suspension 3 days before inoculation with 1 × 106 CFU C. albicans or with 5μL sterile PBS after the corneas were scratched. The mouse corneas or eyeballs were collected at 3 dpi & subjected for further detection. PBMC were isolated from whole blood and stained for surface markers before cells were fixed, permeabilised and stained for TLR-9. FSC and SSC were first used to gate out debris and SSC-A and SSC-H was utilized to eliminate duplicates. Further gating was done on CD45 and CD19, to target B cells. To separate between the different B cell populations, we gated on CD27 and IgD (D), followed by TLR-9 expression on these subsets. Data from one representative patient is shown. THP-1 human acute monocytic leukemia cell line was stained with Mouse anti-TLR9 (26C593.2) Protein-G purified Monoclonal Antibody conjugated to Janelia Fluor® 549 (Catalog # NBP2-24729JF549, blue histogram) or matched control antibody (orange histogram).

Alveolar epithelial cells (AECs), fibroblasts and fibrocytes all express Toll-like receptor-9. AECs, fibroblasts, fibrocytes and whole lungs were isolated from Balb/c, mice and total RNA prepared and analyzed for TLR-9 expression. The data were normalized to the expression level of TLR-9 in dendritic cells, which was set to 1 (n = 3/group). No TLR-9 message was amplified in cells from TLR-9-/- mice. (B, C) Immunohistochemistry was performed on paraffin wax-embedded whole-lung tissue from normal Balb/c mice. (B) Control staining with secondary antibody only; (C) diffuse TLR-9 staining, with notable staining in the bronchial epithelium, at the alveolar junctions and the interstitium. Fibroblasts (E) and AECs (G) were isolated from Balb/c mice and fixed. Immunofluorescence staining for TLR-9 shows that both cell types expressed TLR-9. Western Blot: IL-1 beta/IL-1F2 Antibody [NB600-633] - Antibody was used at a 1:200 dilution incubated 1h at room temperature to detect dog IL-1b by Western blot. Lane 1 and 2 were loaded with 2.5 ug and 500 ng of dog IL-1b respectively. The molecular weight of the detected band is estimated by comparison to molecular weight markers (not shown). Detection occurred using a 1:3,000 dilution of IRDye (TM) 800 conjugated Donkey anti-Rabbit IgG for 1h at room temperature.

Effects of butyrate treatment on curdlan-induced SpA mice. SKG mice were injected with 3 mg of curdlan through intraperitoneal and oral administration. Flow Cytometry: IKK alpha Antibody (14A231) - BSA Free [NB100-56704] - An intracellular stain was performed on Hek293 cells with IKK alpha Antibody (14A231) NB100-56704 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Western Blot: IKK alpha Antibody (14A231) - BSA Free [NB100-56704] - IKK alpha Antibody (14A231) [NB100-56704] - Total protein from various Human, Mouse and Rat cell lines were separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL anti-IKK-alpha in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.

Immunohistochemistry: TLR7 Antibody [NBP2-24906] - Expression of the TLRs 5 and 7 were evaluated in the PDAC tissue by their staining intensity using TLR5 antibody (NBP2-24787) and TLR7 antibody (NBP2-24906). Note negative staining in E and G; positive staining in F and H. For TLR7 immunopositivity was detectable in the cytoplasm with no notable membranous or nuclear positivity. TLR5 showed distinctive nuclear positivity, and detectable positivity in the cytoplasm in some samples. Western Blot: IL-6 Antibody [NB600-1131] - Protein was resolved on a 4-20% Tris-Glycine gel by SDS-PAGE and transferred onto nitrocellulose. The blot shows detection of a band 21 kDa in size corresponding to anti-IL6 antibody. Molecular weight markers are also shown (MW). After transfer, the membrane was blocked for 30 minutes with 1% BSA-TBST. Detection occurred using peroxidase conjugated anti-Rabbit IgG secondary antibody diluted 1:40,000 in blocking buffer for 30 min at RT followed by reaction with FemtoMax chemiluminescent substrate.

Immunocytochemistry/Immunofluorescence: IKK beta Antibody (10AG2) [NB100-56509] - U-87 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. The cells were incubated with anti-IKK beta Antibody (10AG2) at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. 1 ug of antibody was added to 100 uL of staining buffer and cells were incubated for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Flow Cytometry: TLR8 Antibody (44C143) [NBP2-24917] - HEK cells expressing TLR8 were treated with ssRNA40-Alexa 546 followed by fixation, permeablization and staining for TLR8 using anti-TLR8 Alexa 488 conjugate or isotype control IgG1/Kappa Alexa 488. TLR8 detection is shown along the horizontal axis, and ssRNA40 Alexa 546 detection is shown along the vertical axis. Direct excitation (488 and 561 nm lasers) of both fluorophores (Alexa 488 and Alexa 546) allowed quantitation of cells staining positive for TLR8 (anti-TLR8-Alexa488) and cells that had taken up ssRNA40-Alexa 546. Single-stranded RNA40 binds TLR8 (flow cytometry FRET assay). A representative experiment is shown from three independent experiments with similar results.
