![Immunohistochemistry: AIF-1/Iba1 Antibody [NB100-1028] - Temporal activation of caspase-8 in Iba1-positive myeloid cells. Frozen tissue sections from mice upon pMCAO (n = 9) were analyzed by immunohistochemistry for expression of active caspase-8 or for expression of the microglia/macrophage marker Iba1. The contralateral side to the ischemic area was used an internal control for each animal. A small increase in body size of Iba1 positive cells was detected in the peri-infarct area at 6 h, with larger increase in cell body & numbers of cells expressing Iba1 detected at 24 & 48 h post occlusion (a). Cleaved caspase-8 was detected at low levels at 6 h, but a significant increase in expression levels was observed at 24 & 48 h in the peri-infarct area, but not at the contralateral side (a). Double immunofluorescent staining revealed that increased active caspase-8 expression was localized in Iba1-positive cells (b). ![Immunohistochemistry: Goat Polyclonal AIF-1/Iba1 Antibody [NB100-1028] - Pax5 and Iba1 immunoreactivity in a formalin fixed paraffin embedded classic Hodgkin's lymphoma biopsy. Formalin fixed paraffin embedded sections were deparaffinized and rehydrated. Sections then underwent heat induced epitope retrieval in a citrate-based solution for 20min in a food steamer. Sections were cooled, then blocked in phosphate buffered saline containing 0.2% Triton-X100 and 10% normal donkey serum for 1h at room temperature. Sections were then incubated with the primary antibody for 16h at room temperature. The following day slides were washed three times with PBS containing 0.05% Tween-20. Sections were then incubated with donkey anti-goat (647 conjugate) at 5ug per mL for 45min at room temperature. Slides were then washed as above and mounted with a coverslip. PBMC were isolated from whole blood and stained for surface markers before cells were fixed, permeabilised and stained for TLR-9. FSC and SSC were first used to gate out debris and SSC-A and SSC-H was utilized to eliminate duplicates. Further gating was done on CD45 and CD19, to target B cells. Immunofluorescent staining for endothelial cells using an anti‐CD 31 antibody revealed an intact endothelial cell monolayer in the uninjured control carotid artery (N = 3). As expected post balloon angioplasty, there is no endothelial cell monolayer of the injured left carotid arteries in the injury alone, injury + T‐AuNP, & injury + S‐AuNP treatment conditions. Green indicates autofluorescence of the elastic lamina. Magenta indicates endothelial cells. Images obtained using 25× magnification with exposure time of 400 msec. TUNEL staining for apoptosis revealed no evidence of endothelial cell apoptosis in the uninjured control carotid arteries, with a prominent endothelial cell monolayer (white arrowhead). ![Immunohistochemistry-Paraffin: Rabbit Polyclonal CD31/PECAM-1 Antibody [NB100-2284] - CD31 positive vessels in a canine splenic hemangiosarcoma.Tissue slides were loaded into automated research stainer, dewaxed and pretreated with EDTA-based epitope retrieval ER2 solution for 20 mins at 100°C. After a peroxide incubation for 5 min, the rabbit antibody against CD31 (NB100-2284) (1:100) was incubated for 60 mins at room temperature followed by Bond Polymer (anti-rabbit HRP) incubation for 8 min at room temperature. Mixed DAB reagent (Polymer Refine Detection Kit) was incubated for 10 mins, and Hematoxylin (Refine Detection Kit) counterstaining for 10 mins. After staining, sample slides were washed in water, dehydrated using ethanol gradient (70%, 90%, 100%), washed three times, and mounted in mounting medium. ![Western Blot: RIPK3/RIP3 Antibody - BSA Free [NBP1-77299] - Bone-marrow-derived macrophages exhibit significant reduction in RIPK3 levels in Ripk3 delta Global, Ripk3 delta MΦ-Cre & Ripk3 delta EC-Cre mice. (A,B) BMDMs were isolated, differentiated on chamber slides for 7 days, immunostained for macrophage markers CD68 (red; A) & CD11b (pink; B), & co-stained for nuclei (Hoechst; blue). All cells from seven separate BMDM isolations were positive for these markers (n=7). (C) A phagocytosis assay was performed on BMDMs with fluorescent polystyrene beads. Phagocytic cells display TRITC+/Hoechst+ signal, & 85% of cells from two separate BMDM isolations were double positive (n=2). (D-F) Protein or RNA was collected from control, Ripk3 delta Global, Ripk3 delta MΦ-Cre & Ripk3 delta EC-Cre BMDMs. RNA was converted to cDNA & analyzed by qPCR for Ripk3 levels (D). Protein lysates were immunoblotted to identify CD11b, RIPK3 & beta -actin (loading control) (E) & quantified (F). (G) RNA from control & Ripk3 delta Global BMDMs was converted to cDNA & analyzed by qPCR for Ripk3-Exon 10 levels. For panels D,F,G, each dot represents a BMDM isolation from an individual animal. Statistics for panels D & F were calculated using one-way ANOVA with Dunnett's multiple comparisons test. Overall ANOVA P-values (prior to the post hoc tests) are 0.0002 (D) & 76%.
