![Immunohistochemistry-Paraffin: MGMT Antibody (MT 23.2) [NB100-168] - IHC analysis of a formalin fixed paraffin-embedded (FFPE) human kidney using 1:2000 conc. of MGMT antibody (clone MT 23.2) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Nuclear staining of MGMT was observed. (B) The increase in gamma H2AX was also validated through western blot analysis (2.5-fold, p < 0.01, n = 3). ATM phosphorylation on serine1981 is significantly upregulated after knockdown of CAPER expression (5-fold, p < 0.05, n = 3). (C) CAPER knockdown resulted in an increased level of caspase-3/7 activation through decreasing live cells (1.2-fold, p < 0.001, n = 3), while increasing apoptotic (7.5-fold, p < 0.001, n = 3), apoptotic/dead (6.5-fold, p < 0.001, n = 3), & dead (3-fold, p < 0.05, n = 3) cell populations. ![<b>Independent Antibodies Validation. </b>Immunoprecipitation: MDMX Antibody [NB100-556] - Detection of human HdmX/MDM4 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from Jurkat cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-HdmX/MDM4 antibody NB100-556 (lot NB100-556-4) used for IP at 6 ug per reaction. HdmX/MDM4 was also immunoprecipitated by a previous lot of this antibody (lot NB100-556-3) and another rabbit anti-HdmX/MDM4 antibody. For blotting immunoprecipitated HdmX/MDM4, NB100-556 was used at 0.4 ug/ml.
