TIM-4 Antibody (5G3) [CoraFluor™ 1] Summary

• Description: CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

• Immunogen: This antibody was raised by immunising female Lewis rats (Harlan Sprague-Dawley) with Tim-4:Ig fusion protein in complete Freund's adjuvant.
• Specificity: This antibody is specific for murine TIM-4, which unlike other TIM family members, is not expressed on T lymphocytes but rather found on antigen presenting cells such as lymphoid and myeloid DC's and macrophages. TIM-4 has been shown to interact with TIM-1 thereby suggesting a role of TIM-4 in the balance of Th1/Th2 regulation. Additionally TIM-4 has been shown to be a receptor for phosphatidylserine and may play a role in engulfment of apoptotic cells.

Background

The T cell immunoglobulin and mucin domain containing protein (TIM) family encodes cell surface receptors that are involved in the regulation of T helper (Th) -1 and -2 cell-mediated immunity. Studies have shown that TIM 4, which is preferentially expressed on macrophages and dendritic cells, is the natural ligand of TIM 1, and that this binding leads to T-cell expansion and cytokine production. Unlike other members of the TIM family, TIM 4 lacks a putative tyrosine phosphorylation signal sequence in its intracellular domain (Kuchroo et al, 2003). The TIM 4 gene maps to a locus associated with predisposition to asthma in both mice and humans and with its connection to TIM 1-triggered Th2 responsiveness, may be considered as a candidate disease/predisposition gene for asthma
