Antibody: Affinity purified rabbit anti-RAP1 antibody NB100-292 used for WB at 0.1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Antibodies: Affinitypurified rabbit anti-RAP1 antibody used for IP at 6 ug per reaction. RAP1 was also immunoprecipitated by a previous lot of this antibody and rabbit anti-RAP1 antibody. For blotting immunoprecipitated RAP1, was used at 0.4 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background

Mammalian telomeric proteins, including TRF1, TRF2, tankyrase, and TERF21P have no recognized orthologs in budding yeast. However, human Rap1 (hRap1) is an ortholog of the yeast telomeric protein, scRap1p. hRap1 is a homodimer, has three conserved sequence motifs in common with scRap1, is located at telomeres, and plays a role in telomere length regulation. Although scRap1 binds telomeric DNA directly, TERF21P is recruited to telomeres by TERF21P and binds it with its C-terminus, rather than directly binding DNA.
